目的:构建HLAI类抗原的反义逆转录病毒载体以抑制细胞该类抗原的表达。方法:从MBmp18/HLA-B7酶切得HLA-B7cDNA后转染至质粒pGEM-3Z,在酶切得含HLA-B75′端非编码区的818bp片段后,将其反向亚克隆于逆转录病毒pLX5N构建成pLAHLA-B7。结果:经鉴定证明构建的质粒中均显示有目的长度的片段,并证明为反向插入。结论:相应反义逆转录病毒载体的建成,将有助于研究抑制移植物HLAI类抗原的表达。
OBJECTIVE: To construct an antisense retroviral vector containing HLA class I antigens to inhibit the expression of such antigens in cells. METHODS: HLA-B7 cDNA was digested with MBmp18 / HLA-B7 and then transfected into plasmid pGEM-3Z. After digestion with an 818 bp fragment containing the noncoding region of HLA-B75 ’end, it was subcloned into reverse transcription The virus pLX5N was constructed as pLAHLA-B7. Results: All the constructed plasmids were identified as having the purpose of length, and proved to be reverse insertion. Conclusion: The construction of the corresponding antisense retroviral vector will help to study the inhibition of the expression of graft-type HLAI antigens.
