背景 治疗乳腺癌常用大剂量化疗和干细胞移植。肿瘤细胞常存在于干细胞悬液液中,采用CD34+阳性选择除去肿瘤细胞的方法既昂贵又费时。一种有效的、廉价的、易于操作的过滤系统可改善移植前的干细胞制备。方法①除去肿瘤细胞:外周血单个核细胞与低浓度(1-6%)的经培养的乳腺癌细胞混合。每一滤器(10cm~2/滤器,2~4mm厚)滤过5千万个细胞。用流式细胞器测细胞回收和细胞鉴别。②细胞移植:从年青C57BL雄鼠股骨取骨髓细胞,悬浮于RPMI培养液培养。小鼠经致死剂量照射,16小时后经侧尾静脉注入10~5经过滤或未经过滤的细胞。注射后9天杀死小鼠,取出脾脏并固定,计算脾脏克隆
Background Breast cancer is commonly treated with high-dose chemotherapy and stem cell transplantation. Tumor cells are often found in stem cell suspensions. The use of CD34+ positive selection to remove tumor cells is expensive and time consuming. An effective, inexpensive, easy-to-handle filter system improves stem cell preparation prior to transplantation. Method 1 Removal of Tumor Cells: Peripheral blood mononuclear cells were mixed with low concentrations (1-6%) of cultured breast cancer cells. Each filter (10cm~2/filter, 2~4mm thick) filters over 50 million cells. The cell recovery and cell identification were measured using a flow cytometer. 2 Cell transplantation: Bone marrow cells were collected from the femur of young C57BL males and suspended in RPMI medium. The mice were exposed to lethal doses and 16 to 10 hours later they were injected with 10 to 5 filtered or unfiltered cells via the lateral tail vein. 9 days after the injection, the mice were killed, the spleen was removed and fixed, and the spleen clone was counted.
