目的探讨三七总皂苷(Panax Notoginseng Saponins,PNS)对肺纤维化小鼠肺组织蛋白酶K(cathepsin K,CatK)表达的影响。方法 C57BL/6雄性小鼠90只,分为假手术组、模型组和PNS组,每组30只。除假手术组外其余各组小鼠气管内一次性滴注盐酸博莱霉素(Bleomycin,BLM),假手术组小鼠气管内一次性滴注等体积的生理盐水。造模后第2天开始给药,持续到处死动物的前1天。假手术组、模型组分别灌服等体积的生理盐水。各组动物于7,14,28 d随机取10只摘眼球取血,分离血清,采用放免法检测血清中层粘连蛋白(LN)和透明质酸(HA)的含量;并处死取肺组织进行HE染色,免疫组织化学法和Western-blot法检测CatK蛋白表达情况。结果 PNS组血清中LN和HA的含量较模型组明显降低(P<0.01或P<0.05),肺泡炎及纤维化程度较模型组明显减轻,CatK的表达升高(P<0.01或P<0.05)。结论 PNS对BLM诱导的小鼠肺纤维化有一定的防治作用,其作用机制可能与上调CatK的表达有关。
Objective To investigate the effect of Panax Notoginseng Saponins (PNS) on the expression of cathepsin K (CatK) in lung tissue of mice with pulmonary fibrosis. Methods Ninety male C57BL / 6 mice were divided into sham operation group, model group and PNS group, 30 rats in each group. Except the sham-operation group, the mice in other groups were given a single instillation of Bleomycin Hydrochloride (BLM) in the trachea, and a single intraperitoneal instillation of normal saline in sham operation group. Dosing began on the second day after modeling and lasted one day before the animal was sacrificed. Sham operation group, model group were fed with equal volume of normal saline. At 7, 14, and 28 days, 10 rabbits in each group were randomly selected to take the blood and the serum was separated. The contents of serum laminin (LN) and hyaluronic acid (HA) were determined by radioimmunoassay. Staining, immunohistochemistry and Western-blot assay CatK protein expression. Results The levels of LN and HA in PNS group were significantly lower than those in model group (P <0.01 or P <0.05), and the degree of alveolitis and fibrosis were significantly reduced compared with model group (P <0.01 or P <0.05) ). Conclusion PNS can prevent and cure BLM-induced pulmonary fibrosis in mice. The mechanism may be related to the up-regulation of CatK expression.
