目的:通过6种中药对小鼠的细胞因子受体影响探索四气药性的免疫分子机理。方法:SD小鼠随机分正常对照组、附子组、干姜组、肉桂组、黄连组、栀子组、知母组。6种中药的水浓缩煎剂每天分别对各自的给药组小鼠灌胃1次,连续10d,按照制定的实验动物中医证候体征级别计分评定观察小鼠情况,10d小鼠眼球取血,用Luminex x MAP technology液相悬浮芯片技术对小鼠血浆进行可溶性CD30(s CD30)、可溶性白介素-1受体Ⅰ(s IL-1RⅠ)、可溶性白介素-2受体α(s IL-2Rα)、可溶性白介素-4受体(s IL-4R)、可溶性白介素-6受体(s IL-6R)等可溶性细胞因子受体指标的定量测定。结果:附子组的小鼠总进食量显著减少;肉桂组的小鼠总饮水量显著增加;各给药组小鼠胃粘膜出血溃疡显著增加;附子组的小鼠s CD30显著增加;肉桂组的小鼠s IL-1R1显著增加;附子组肉桂组的小鼠s IL-2Rα显著增加;黄连组的小鼠s IR-4R显著增加;附子组干姜组肉桂组黄连组的小鼠s IL-6R显著增加。结论:为受试中药药性的细胞因子受体信号转导表征免疫分子机理探索提供中医理论的实验基础。
OBJECTIVE: To explore the immune molecular mechanism of four gas herbs by the influence of six kinds of traditional Chinese medicine on cytokine receptors in mice. Methods: SD mice were randomly divided into normal control group, aconite group, dried ginger group, cinnamon group, Coptis group, Zhizi group, Zhimu group. Six kinds of Chinese medicine water concentrated decoction were administered to mice in their respective administration groups once a day for 10 consecutive days. The mice were observed and scored according to the TCM syndrome rating scale of the experimental animals. Blood was collected from the eyeballs of the mice on the 10th day The soluble CD30 (sCD30), sIL-1RⅠ, sIL-2Rα, sIL-1Rα and sIL-1Rα in mouse plasma were detected by Luminex x MAP technology. , Soluble interleukin-4 receptor (sIL-4R), soluble interleukin-6 receptor (sIL-6R) and other soluble cytokine receptor. Results: The total food intake of aconite group was significantly decreased; the total water consumption of mice in cinnamon group was significantly increased; gastric mucosal hemorrhage and ulcer of mice in each group were significantly increased; sCD30 of aconite group was significantly increased; SIL-1R1 increased significantly in mice in the aconite group and s IR-2Rα in mice in the rhizoma coptidis group increased significantly 6R significantly increased. Conclusion: This study provides an experimental basis for exploring the molecular mechanism of cytokine receptor signaling transduction in Chinese medicine.
