目的:观察痛泻要方拆分成分防风(RS)、白术(LA)和白芍(WPR)对离体大鼠结肠平滑肌的影响,并探讨其部分机制。方法:制备大鼠结肠平滑肌肌条,以9g·L-1NaCl溶液为对照,观察白术、白芍和防风对平滑肌自发收缩的影响和防风对乙酰胆碱(Ach)引起的肌条收缩的影响,并用3种工具药(纳络酮(NLT)、心得安(XDA)、酚妥拉明(FILM))初步探讨了防风的作用机制。结果:与NS组相比较,白术、白芍均对大鼠平滑肌有促进收缩的作用,且白术的作用大于白芍;防风各剂量组(10~40g·L-1)均对平滑肌收缩有剂量依赖性的抑制作用。不同浓度的防风对Ach引起的肌条收缩作用均有抑制,并呈剂量依赖关系。先用工具药充分孵育后再加入RS,与对照组(NS+RS)比较,NLT+RS组面积的回升有统计学意义(P<0.05);FTLM组振幅和面积回升差异均有统计学意义(P<0.01),与阴性对照组(NS)的差异则无统计学意义。结论:痛泻要方主要成分白术和白芍对大鼠结肠平滑肌均有促进收缩的作用。防风有抑制作用,其作用机制与肾上腺素能α受体有关,与胆碱能M受体可能也有一定关系。
OBJECTIVE: To observe the effects of the components of TongxieYao prescription on the isolated rat colonic smooth muscle cells (RS), Atractylodes macrocephala (LAR) and white peony root (WPR), and discuss some of its mechanisms. Methods: The rat colon smooth muscle strips were prepared and the effect of Atractylodes, Radix Paeoniae Alba and windproof on spontaneous contraction of smooth muscle and the contractions of muscle strips caused by acetylcholine (Ach) were observed with 9g · L-1 NaCl solution. (NLT, XDA) and phentolamine (FILM) were used to explore the mechanism of windbreak. Results: Compared with NS group, Atractylodes macrocephalae and Radix Paeoniae Alba were effective in promoting smooth muscle contraction of rats and Atractylodes macrocephala was more effective than Radix Paeoniae Alba. Radix et Rhizoma Rhei was effective in contracting smooth muscle cells in doses of 10 ~ 40g · L-1 Dependence of the inhibitory effect. Different concentrations of windproof Ach induced contraction of muscle strips were inhibited, and in a dose-dependent manner. RSV was increased after treatment with the drug, and the area of NLT + RS increased significantly (P <0.05) compared with the control group (NS + RS). The amplitude and area recovery of FTLM group were statistically significant (P <0.01), but no significant difference with negative control group (NS). Conclusion: The main ingredients of Tong Xie Yao Fang and Radix Paeoniae Alba are able to promote contraction of rat colonic smooth muscle. Windproof inhibition, its mechanism of action and adrenergic receptor α, and cholinergic M receptor may also have a certain relationship.
