目的探讨在小鼠淋巴道转移潜能不同的肝癌细胞中,肝癌细胞增殖侵袭能力与膜联蛋白A7的亚细胞定位和亚型的表达。方法以小鼠腹水型肝癌高、低淋巴道转移潜能不同的细胞株Hca-F细胞和Hca-P细胞为研究对象,通过CCK8法检测细胞的增殖能力,Transwell小室检测细胞侵袭能力;Western blotting方法检测膜联蛋白A7在体外增殖侵袭能力不同的肿瘤细胞中的亚细胞定位和亚型的变化。结果发现Hca-F细胞无论是增殖能力还是侵袭能力都高于Hca-P细胞。膜联蛋白A7在Hca-F细胞和Hca-P细胞主要为47 kD亚型表达,而51 kD亚型只少量表达于细胞浆。47 kD亚型在Hca-F细胞的细胞浆、细胞膜、细胞器膜以及细胞骨架中的表达均明显高于Hca-P细胞,分别为(73%±7%)v(s49%±19%)、(36%±8%)v(s24%±9%)和(53%±7%)v(s40%±12%)(均P<0.05)。结论膜联蛋白A7的47 kDa亚型在细胞浆、细胞膜、细胞器膜和细胞骨架的定位表达,可能与肝癌细胞增殖侵袭能力密切相关。
Objective To investigate the expression of subcellular localization and subtype of Annexin A7 in hepatocellular carcinoma cells in the proliferation and invasion of hepatoma cells in mice with different lymphatic metastatic potential. Methods Hca-F cells and Hca-P cells with different high and low lymphatic metastatic potential of mouse ascites hepatocarcinoma were used as research objects. Cell proliferation was detected by CCK8 assay. Transwell chambers were used to detect cell invasion. Western blotting To detect subcellular localization and subtype changes of annexin A7 in tumor cells with different invasive and proliferative capacities in vitro. The results showed that Hca-F cells were higher than invasive or invasive ability of Hca-P cells. Annexin A7 was mainly expressed in the 47 kD subtype of Hca-F cells and Hca-P cells, whereas the 51 kD subtype was only expressed in the cytoplasm in small quantities. The expression of 47 kD subtypes in Hca-F cells was significantly higher than that in Hca-P cells (73% ± 7%) (49% ± 19%) in the cytoplasm, cell membrane, organelle membrane and cytoskeleton, (36% ± 8%) v (s24% ± 9%) and (53% ± 7%) v (s40% ± 12%) (all P <0.05). Conclusion Localization of the 47 kDa subunit of Annexin A7 in cytoplasm, membrane, organelle membrane and cytoskeleton may be closely related to the proliferation and invasiveness of hepatocellular carcinoma cells.
