目的 :体外探讨乳猪与幼鲨肝细胞生长因子 (HGF)对人细胞因子分泌的影响 ,了解HGF的作用机制及其对机体免疫系统的调节作用。方法 :将人外周血单个核细胞 (PBMC)悬液与HGF共同孵育诱导后 ,分别采用酶联免疫吸附试验和细胞学技术检测IL 2 ,IFNγ与TNFβ的分泌水平。结果 :鲨鱼HGF能明显促进淋巴细胞分泌IFNγ(P <0 .0 1 ) ,抑制TNFβ的表达(P <0 .0 1 ) ,但不影响IL 2的表达水平。乳猪HGF能有效抑制TNFβ的分泌 (P <0 .0 1 ) ,对IFNγ和IL 2的分泌则无明显的促进作用。结论 :HGF尤其是鲨鱼HGF ,可有效调节淋巴因子的分泌水平 ,提示其在改善机体免疫调节及免疫防御机制等方面具有一定的研究与应用价值。
OBJECTIVE: To investigate the effects of hepatocyte growth factor (HGF) on the secretion of human cytokines in suckling pigs and young shark in vitro, and to understand the mechanism of action of HGF and its regulation on the immune system. Methods: Human peripheral blood mononuclear cells (PBMC) suspension were incubated with HGF for inducing. The levels of IL 2, IFNγ and TNFβ were detected by enzyme-linked immunosorbent assay (ELISA) and cytology respectively. Results: Shark HGF could significantly promote the secretion of IFNγ by lymphocytes (P <0.01) and inhibit the expression of TNFβ (P <0.01), but did not affect the expression of IL-2. The piglets HGF can effectively inhibit the secretion of TNFβ (P <0.01), but not the secretion of IFNγ and IL 2. CONCLUSION: HGF, especially shark HGF, can effectively regulate the secretion of lymphokines, suggesting that it has certain research and application value in improving the immune regulation and immune defense mechanism.
