目的:探讨喉癌中MDM2和p16基因的表达与喉病变组织的关系,并对MDM2和p16基因在喉癌中表达的关系进行初步研究。方法:应用鼠抗人MDM2单克隆抗体和鼠抗人p16单抗,采用SP免疫组织化学对35例喉鳞状细胞癌(LSCC)、21例喉不典型增生、20例正常喉组织中MDM2蛋白和p16蛋白进行检测。结果:癌周正常喉组织、不典型增生组织和LSCC组织中MDM2蛋白的表达率分别为10.0%、38.1%和54.3%(P<0.05),MDM2蛋白阳性表达率在病理Ⅰ~Ⅲ级肿瘤之间分别为38.1%、66.7%、100.0%,呈明显升高趋势(P<0.05);p16在喉正常组织、不典型增生组织和LSCC组织中的阳性表达率分别为85.1%、52.4%、34.3%(P<0.05),在Ⅰ~Ⅲ级LSCC中的表达分别为52.4%、11.1%和0(P<0.05);MDM2蛋白表达与p16蛋白表达呈负相关(P<0.05)。结论:MDM2蛋白的表达阳性率伴随组织分化程度降低而明显提高,对LSCC诊断有重要辅助意义;p16蛋白的表达伴随组织分化程度的降低而明显降低,在低分化LSCC中不表达,这对于判断LSCC预后有重要意义;MDM2和p16呈相反的表达趋势,两者呈现一定程度的负相关。
Objective: To investigate the relationship between the expression of MDM2 and p16 in laryngeal carcinoma and laryngeal lesions, and to study the relationship between the expression of MDM2 and p16 in laryngeal carcinoma. Methods: Mouse anti-human MDM2 monoclonal antibody and mouse anti-human p16 monoclonal antibody were used to detect the expression of MDM2 protein in 35 cases of laryngeal squamous cell carcinoma (LSCC), 21 cases of atypical hyperplasia of the larynx and 20 cases of normal laryngeal tissues by SP immunohistochemistry. And p16 protein were detected. Results: The expression rates of MDM2 protein in normal laryngeal tissues, atypical hyperplasia tissues and LSCC tissues were 10.0%, 38.1% and 54.3%, respectively (P <0.05). The positive rates of MDM2 protein in pathological grade Ⅰ ~ Ⅲ tumors (P <0.05). The positive rates of p16 expression in normal laryngeal tissues, atypical hyperplasia tissues and LSCC tissues were 85.1%, 52.4% and 34.3 %, Respectively (P <0.05). The expression of MDM2 protein was negatively correlated with the expression of p16 protein in grade Ⅰ ~ Ⅲ LSCC (P <0.05). CONCLUSION: The positive rate of MDM2 protein expression is significantly decreased with the decrease of tissue differentiation, which is of important adjuvant significance for the diagnosis of LSCC. The expression of p16 protein is decreased with the decrease of tissue differentiation and is not expressed in poorly differentiated LSCC, The prognosis of LSCC is of great significance. MDM2 and p16 show the opposite trend of expression, showing a certain degree of negative correlation.
