目的建立一种快速灵敏测定尿样中苯代谢物氢醌和邻苯二酚含量的高效液相色谱-电化学检测方法(HPLC-ECD)。方法采用ODS柱,流动相为pH 5.84缓冲液(0.01 mol/L柠檬酸三钠和0.01 mol/L磷酸氢二钠:乙腈(V/V)=850:150的溶液,流速1.0 ml/min;结合气质联用技术,复核结果的可靠性。结果氢醌和邻苯二酚线性范围分别为1.25~100.20 ng/ml和2.50~125.20 ng/ml,氢醌和邻苯二酚回收率分别为92.3%~102.4%和90.6%~106.2%,氢醌和邻苯二酚检出限的为0.05 ng/ml。结论方法简单、快速、灵敏,可用于尿样中苯代谢物氢醌和邻苯二酚的检测。
OBJECTIVE To establish a rapid and sensitive HPLC-ECD method for the determination of hydroquinone and catechol in benzene metabolites in urine samples. Methods ODS column was used. The mobile phase consisted of a solution of 0.01 mol / L sodium citrate and 0.01 mol / L disodium hydrogen phosphate: acetonitrile (V / V) = 850: 150 with a flow rate of 1.0 ml / min. Results The reliability of hydroquinone and catechol ranged from 1.25 to 100.20 ng / ml and from 2.50 to 125.20 ng / ml, respectively. The recoveries of hydroquinone and catechol were 92.3 % ~ 102.4% and 90.6% ~ 106.2%, respectively.The detection limits of hydroquinone and catechol were 0.05 ng / ml.Conclusion The method is simple, rapid and sensitive and can be used for the determination of benzene hydroquinone and o- Phenol detection.
