目的:为了探索大面积Ⅲ度烧伤混合植皮创面愈合过程中生长因子的作用。方法:本实验以大鼠为模型,在异体皮移植后3、5、7、14、21和28天以及自体皮移植后2、4、11、18和25天分别取自体皮区、异体皮区皮肤,用酸性硫氰酸胍苯酚氯仿(AGPC)法提取总RNA,经逆转录聚合酶链反应(PTPCR),测定TGFα、TGFβ1、PDGF和bFGF的mRNA表达量。结果:1)这四种生长因子mRNA表达在自体皮区及异体皮区各时相都能被检测到,并且自体皮区表达均高于异体皮区。2)在自体皮区,TGFα的表达在2大以后持续升高,表达峰值在18天;TGFβ1在2天就有高表达,第二峰值出现在18天;PDGF在2天有较高表达,表达峰值在4天,随后逐渐下降,25天回升:bFGF也在2天有很高表达,以后下降,象PDGF一样,25天回升。3)在异体皮区,创面愈合早期这四种生长因子表达都较低,以后各有不同程度升高。结论:1)移植的自体皮与异体皮都存活,并反映了自体皮区的愈合再生较异体皮区旺盛。2)在自体皮区这四种生长因子通过自分泌作用,旁分泌作用,到达创伤区,沿着信号转导通路,相互协同发挥效应,促进创面愈合。3)在异体皮区存在排异反应,?
Objective: To explore the role of growth factors in wound healing of mixed skin grafts with large area Ⅲ degree burns. Methods: The rats were used as the model and were harvested at 3, 5, 7, 14, 21 and 28 days after allograft skin transplantation and at 2, 4, 11, 18 and 25 days after autologous skin graft, The total RNA was extracted by acid guanidinium thiocyanate (AGPC) method and the mRNA expression of TGFα, TGFβ1, PDGF and bFGF was determined by reverse transcription-polymerase chain reaction (PTPCR). Results: (1) The mRNA expression of these four growth factors could be detected in autophagic and allogeneic dermatomes, and the expression of autologous skin was higher than that of allogeneic dermis. 2) In the autologous skin area, the expression of TGFα continued to increase after 2 days, the expression peak was at 18 days. TGFβ1 was highly expressed at 2 days and the second peak appeared at 18 days. PDGF was highly expressed at 2 days, The peak of expression was at 4 days, then decreased gradually and rebounded at 25 days: bFGF was also highly expressed in 2 days and then decreased. Like PDGF, bFGF rose again in 25 days. 3) The expression of these four growth factors was lower in allogeneic skin and early wound healing, and each increased in different degrees afterwards. Conclusions: 1) Both autologous skin and allogeneic skin all survived, which reflected that the healing of autologous skin area was more plentiful than that of allogenic skin area. 2) In the autologous skin area, these four growth factors reach the wound area through autocrine and paracrine effects and work along the signal transduction pathways to synergize with each other to promote wound healing. 3) there is rejection in allogeneic skin,?
