以脂质体为载体,把突变的抗MTX的耐药基因转染到小鼠的造血干细胞中,用不同浓度的MTX对细胞进行筛选。结果发现同一浓度下已转染基因组的细胞接种率大于未转染基因组的细胞接种率。未转染组对MTX耐受性差,克隆形成率低。行卡方检验,同一浓度下的组间差异显著(P<0.001)。PCR分析证实抗MTX的耐药基因在造血于细胞中有效表达。该实验为进一步研究MTX治疗的肿瘤患者减少骨髓毒性,提供了有参考价值的资料。
The mutated anti-MTX gene was transfected into mouse hematopoietic stem cells using liposome as a vector, and the cells were screened with different concentrations of MTX. As a result, it was found that the cell inoculation rate of the transfected genome was greater than the cell inoculation rate of the untransfected genome at the same concentration. The untransfected group had poor tolerance to MTX and low colony formation rates. In the chi-square test, there was a significant difference between groups at the same concentration (P<0.001). PCR analysis confirmed the effective expression of anti-MTX resistance genes in hematopoietic cells. This experiment provided valuable reference data for the further study of bone marrow toxicity in patients with MTX therapy.
