本文叙述用放射免疫测定法(RIA)测定毛花洋地黄(Digitalis lanata)不同器官组织中的强心甾含量。已知RIA法中以~3H-狄戈辛甙元(~3H-di-goxigenin)作为示踪物可检测低至0.5ng/试验的狄戈辛甙元衍生的强心甾。所选择的抗血清显示与洋地黄毒甙元和芰毒甙元衍生物有一种显著相关的交叉反应。由于非强心甾有高度的分辨力,所以测定前没有必要对植物提取物进行提纯。所得结果以狄戈辛当量(DE)表示。从狄戈辛溶液与植物提取物得到的稀释曲线作比较,可用以证明缺乏与某种抗体结合的强心甾,
This article describes the determination of the cardiotoxin content in different organs of Digitalis lanata by radioimmunoassay (RIA). It has been known that as low as 0.5 ng / dose of digoxin aglucone derived cardiotonic is detectable with ~ 3H-di-goxigenin as a tracer in the RIA method. The selected antisera showed a significant correlation with cross-reaction with digoxigenin and toxin derivatives. Because of the high resolution of non-cardiac stanols, it is not necessary to purify plant extracts prior to measurement. The results are expressed in terms of Dioxin equivalent (DE). A comparison of the dilution curves obtained from the solution of Digoxigen and plant extracts can be used to demonstrate the lack of a cardiotoxin that binds to an antibody,
