目的 研究羟基喜树碱 (HCPT )对家兔环孢素A(CsA)药物动力学及对红细胞膜流动性 (LFU)的影响。方法 用TDX快速测定CsA全血药物浓度 ,用DPH荧光探针法测定红细胞膜流动性。结果 HCPT可使CsA的a ,V(c) ,K12 及CL(s)降低 ,T1/ 2 (a) 增加 (P <0 0 5 )。联合给药组的CsA血药浓度在给药后的 0 2 5 ,0 5和 1h高于单独给药组 ,且在 1h时差异有显著性 (P <0 0 5 )。联合给药组的LFU值在给药后的 0 5 ,1 0 ,1 5h时高于单独给药组 (P <0 0 5 ) ,与对照组差异无显著性。单独给药组的LFU值在给药后的 0 5 ,1 0 ,1 5h时低于对照组。结论 HCPT与CsA合用可提高CsA的血药浓度 ,有一定的临床应用前景
Objective To study the effects of HCPT on the pharmacokinetics of cyclosporin A (CsA) in rabbits and on the membrane fluidity (LFU) of erythrocytes. Methods Rapid determination of CsA whole blood drug concentration by TDX and determination of erythrocyte membrane fluidity by DPH fluorescence probe method. Results HCPT decreased a, V (c), K12 and CL (s) of CsA and increased T1 / 2 (a) (P <0 05). The concentration of CsA in the combination group was higher than that in the control group at 0 2 5, 0 5 and 1 h after administration, and the difference was significant at 1 h (P <0 05). The LFU value of the combination group was higher than that of the drug alone group (P <0 05) at 0 5, 1 0, 1 5 h after the administration, and there was no significant difference with the control group. LFU value of the group administered alone was lower than that of the control group at 0 5, 1 0, 1 5 h after administration. Conclusion The combination of HCPT and CsA can increase the blood concentration of CsA, and has certain clinical application prospect
