为探讨牛种布鲁氏菌sigma因子RpoE2的功能,采用酵母双杂交技术对RpoE2蛋白与anti-sigma因子之间的互作进行研究;通过环境应激试验评价了rpoE2缺失株对强氧化环境、酸性pH、杀菌性阳离子多肽、铁缺乏环境的耐受能力;通过比较亲本菌株和缺失株在巨噬细胞感染模型和小鼠感染模型中的生存能力,研究了rpoE2基因是否与牛种布鲁氏菌毒力相关。结果显示:牛种布鲁氏菌的RpoE2蛋白可以与anti-sigma因子相互作用,牛种布鲁氏菌rpoE2基因缺失后,对各种应激环境的耐受能力均无影响,并且巨噬细胞和小鼠感染模型中毒力均不降低。牛种布鲁氏菌的RpoE2蛋白是一个ECF16家族的sigma因子,并且其与布鲁氏菌的毒力无关。
In order to explore the function of sigma factor RpoE2 in Brucella bovina, yeast two-hybrid technique was used to study the interaction between RpoE2 protein and anti-sigma factor. Environmental stress tests were carried out to evaluate the effect of rpoE2 deletion on strong oxidative environment, Acid pH, bactericidal cationic polypeptide and iron-deficient environment. By comparing the viability of the parent strain and the deletion strain in the macrophage infection model and the mouse infection model, whether the rpoE2 gene and the bovine Brucella Virulence related. The results showed that the RpoE2 protein of Brucella bovin could interact with anti-sigma factor, and the absence of rpoE2 gene of Brucella bovina caused no effect on the tolerance of various stress conditions. And the macrophage And mice infected with the model does not reduce the virulence. The RpoE2 protein from Brucella bovinus is a sigma factor of the ECF16 family and is not associated with the virulence of Brucella.
