目的研究抵抗素基因外显子4第1308的位点核苷酸G/A变异与武汉地区2型糖尿病和肥胖者发病风险的关系。方法应用聚合酶链反应-限制性片段长度多态性方法检测武汉地区汉族160例抵抗素基因外显子4第1308位点的G/A变异。结果2型糖尿病组和肥胖组的GG、AA基因型及G/A等位基因频率与正常组比较,均有显著性差异(P<0.05)。GG基因型携带者使个体患肥胖和糖尿病的危险性分别增加1.33倍和2.22倍,将G等位基因视为暴露因素,其使个体患肥胖以及糖尿病的危险性分别增加1.12倍和1.80倍。结论抵抗素基因外显子4第1308位点核苷酸多态性与2型糖尿病和肥胖的易感性相关,等位基因G可能是由肥胖致2型糖尿病的一个重要的相关因素。
Objective To investigate the relationship between nucleotide G / A mutation at position 1308 of exon 4 of resistin gene and the risk of type 2 diabetes and obesity in Wuhan. Methods G / A mutation of 1608 exon 4 in Wuhan Han population was detected by polymerase chain reaction - restriction fragment length polymorphism (PCR - RFLP). Results The frequencies of GG, AA genotype and G / A allele in type 2 diabetes mellitus and obesity group were significantly different from those in normal group (P <0.05). GG genotype carriers increased their risk of obesity and diabetes by 1.33 times and 2.22 times, respectively. Using the G allele as an exposure factor increased individual risk of obesity and diabetes by 1.12 times and 1.80 times, respectively. Conclusion The nucleotide polymorphism of 1308 in exon 4 of resistin gene is associated with the susceptibility to type 2 diabetes and obesity. Allele G may be an important factor related to type 2 diabetes caused by obesity.
