目的探讨白黎芦醇对人皮肤瘢痕组织成纤维细胞增殖及Ⅰ、Ⅲ型胶原合成及降解影响的机理。方法用MTT法检测及原位杂交技术,对增生性瘢痕用白黎芦醇作用不同时间后,成纤维细胞的增殖及Ⅰ、Ⅲ型前胶原mRNA的原位表达进行了研究。结果(1)白黎芦醇对人皮肤瘢痕组织成纤维细胞的抑制率随浓度的增加和作用时间的延长而增强,最大抑制率可达到93.69%。(2)Ⅰ、Ⅲ型前胶原mRNA 3 d已被明显抑制(P<0.01),7 d后表达强度进一步下降,白黎芦醇对Ⅰ型前胶原mRNA的抑制作用要比Ⅲ型前胶原强。结论(1)白黎芦醇对人皮肤瘢痕组织成纤维细胞增殖有抑制作用,且呈剂量时间依赖性。(2)白黎芦醇对Ⅰ、Ⅲ型前胶原mRNA有明显抑制作用,对Ⅰ型前胶原mRNA的抑制作用要比Ⅲ型前胶原强。
Objective To investigate the mechanism of resveratrol on the proliferation of human skin scar tissue fibroblasts and the synthesis and degradation of type I and type III collagen. Methods Using MTT assay and in situ hybridization technique, the proliferation of fibroblasts and the in situ expression of type I and type III procollagen mRNAs were studied after different time with the effect of resveratrol on hypertrophic scars. Results (1) The inhibitory rate of resveratrol on fibroblasts of human skin scars increased with the increase of concentration and the prolongation of action time, and the maximum inhibition rate was 93.69%. (2) Type I and III procollagen mRNA was significantly inhibited for 3 days (P<0.01), and the expression intensity was further decreased after 7 days. The inhibitory effect of resveratrol on type I procollagen mRNA was stronger than that of type III procollagen. . Conclusion (1) Resveratrol can inhibit the proliferation of human skin scar tissue fibroblasts in a time-dependent manner. (2) Resveratrol had significant inhibitory effects on type I and type III procollagen mRNA, and its inhibitory effect on type I procollagen mRNA was stronger than that of type III procollagen.
