建立了竹叶中总多酚含量的F olin-Ciocalteu比色测定方法。以没食子酸为对照品,对竹叶中总多酚含量测定的比色条件,包括检测波长、碳酸钠溶液用量、Folin-Ciocalteu试剂用量、反应温度及时间对吸光度的影响进行了研究。结果表明:在0.944—7.552μg/mL范围内,对照品校准曲线为y=0.1237x+0.0469,R2=0.9991,线性关系良好。优化的测定方法为样品提取液0.5mL,F olin-Ciocalteu试剂1.0mL、16%碳酸钠溶液4.0m L、30℃避光反应1.5h,此条件下平均回收率为105.2%(RSD=4.225%)。采用建立的检测方法,测定了12种竹叶中总多酚含量,结果表明竹叶中总多酚平均含量为3.265mg/g;含量最高为龙竹6.059mg/g,最低为佛肚竹0.968mg/g。该方法简便快速、结果准确、重现性和稳定性好,可用于竹叶中总多酚含量的测定。
F olin-Ciocalteu colorimetric method was established for total polyphenol content in bamboo leaves. Colorimetric conditions, including the detection wavelength, the amount of sodium carbonate solution, the amount of Folin-Ciocalteu reagent, reaction temperature and time, on the absorbance of gallic acid were compared with gallic acid as control. The results showed that the calibration curve of the standard was y = 0.1237x + 0.0469 and R2 = 0.9991 in the range of 0.944-7.552μg / mL with good linearity. The optimized method was 0.5 mL of sample extract, 1.0 mL of Folin-Ciocalteu reagent, 4.0 mL of 16% sodium carbonate solution and 1.5 h of light at 30 ° C. The average recovery was 105.2% (RSD = 4.225% ). The results showed that the total content of total polyphenols in bamboo leaves was 3.265mg / g, the highest content was 6.059mg / g, while the lowest was 0.668 mg / g. The method is simple, rapid, accurate, reproducible and stable and can be used for the determination of total polyphenols in bamboo leaves.
