目的 :探讨通脉宁胶囊 (TMNJN )对家兔颈内动脉内皮球囊损伤后血管狭窄模型血液流变学及相关活性因子的影响。方法 :在无菌条件下分离出颈内和颈外动脉 ,由颈外动脉向颈内动脉插入气囊 ,在 2 pa恒定压力下 ,反复抽拉3次 ,造成经腔内皮损伤和血管狭窄。连续给药 2 1天后 ,颈总动脉取血 ,分别测定各实验组家兔全血黏度、血浆黏度、血球压积、红细胞聚集指数、聚集面积、红细胞变形指数、变形面积及血浆血栓素 (TXB2 )、6-酮 (6-keto -PGF1α)含量的变化。结果 :同假手术组相比 ,模型组家兔在造型 2 1天后全血黏度、红细胞压积、红细胞聚集指数、血浆TXB2 、TXB2 / 6-keto -PGF1α含量及比值明显升高 ,6-keto -PGF1α、红细胞变形指数明显降低。同模型组相比 ,造型家兔灌服通脉宁胶囊后 ,大、中剂量组全血黏度、红细胞压积、红细胞聚集指数、TXB2 、TXB2 / 6-keto -PGF1α明显降低 ,6-keto -PGF1α、红细胞变形指数明显升高。结论 :通脉宁胶囊具有较强的活血化瘀作用 ,为通脉宁胶囊能治疗血管内皮球囊损伤引起的血管狭窄提供了充实的药理学基础。
Objective: To investigate the effect of Tongmaining Capsule (TMNJN) on hemorheology and related active factors in vascular stenosis model of rabbits with internal carotid artery endothelial balloon injury. METHODS: The internal and external carotid arteries were isolated under aseptic conditions. The external carotid artery was inserted into the internal carotid artery and the balloon was repeatedly pulled 3 times under a constant pressure of 2 Pa, resulting in transluminal endothelial injury and vascular stenosis. After 21 days of continuous administration, blood was drawn from the common carotid artery to determine the rabbit’s whole blood viscosity, plasma viscosity, hematocrit, erythrocyte aggregation index, aggregation area, red blood cell deformation index, deformation area, and plasma thromboxane (TXB2) in each experimental group. ), Changes in 6-keto (6-keto-PGF1α) content. RESULTS: Compared with the sham-operated group, the blood viscosity, hematocrit, erythrocyte aggregation index, plasma TXB2, TXB2/6-keto-PGF1α content and ratio of rabbits in the model group were significantly increased after 2 days of modeling, 6-keto - PGF1α, erythrocyte deformability index was significantly reduced. Compared with the model group, after the model rabbits fed Tongmaining capsule, the blood viscosity, hematocrit, erythrocyte aggregation index, TXB2, and TXB2/6-keto-PGF1α were significantly reduced in the large and middle dose groups, 6-keto- PGF1α and erythrocyte deformability index increased significantly. Conclusion: Tongmaining Capsule has a strong effect on promoting blood circulation and removing blood stasis. It provides a substantial pharmacological basis for Tongmaining Capsule to treat vascular stenosis caused by vascular endothelial balloon injury.
