目的研究胃癌组织中smad4mRNA、转化生长因子β1(TGFβ1)、转化生长因子β1受体(TGFβR1)的表达特征及其意义。方法常规石蜡包埋切片行smad4mRNA原位杂交染色及TGFβ1、TGFβR1免疫组织化学染色。结果smad4mRNA、TGFβ1、TGFβR1在胃腺癌、黏液腺癌和印戒细胞癌中阳性率无明显差异;组织学分级Ⅰ级、癌细胞未浸润至深肌层和淋巴结未转移的组织smad4mRNA、TGFβ1、TGFβR1阳性率明显高于组织学分级Ⅲ级、癌细胞浸润至深肌层或浆膜层和淋巴结转移组织,差异均有显著或非常显著性意义;smad4mRNA、TGFβ1、TGFβR1在胃癌中表达密切相关。结论smad4mRNA、TGFβ1和TGFβR13者表达与胃癌发生发展、生物学行为和预后可能有关,可作为重要的生物学标记物。
Objective To investigate the expression and significance of smad4 mRNA, transforming growth factor-β1 (TGFβ1) and transforming growth factor-β1 receptor (TGFβR1) in gastric cancer. Methods Paraffin-embedded sections were stained by in situ hybridization for smad4 mRNA and immunohistochemical staining for TGFβ1 and TGFβR1. Results The positive rates of smad4 mRNA, TGFβ1 and TGFβR1 in gastric adenocarcinoma, mucinous adenocarcinoma, and signet-ring cell carcinoma were not significantly different; smad4 mRNA, TGFβ1, TGFβR1 were not histologically graded, and the tumor cells were not infiltrated to the deep myometrium and lymph nodes. The positive rate was significantly higher than histological grade III, cancer infiltration to deep muscle or serosal layer and lymph node metastasis. The differences were significant or very significant; smad4 mRNA, TGFβ1, TGFβR1 were closely related to gastric cancer. Conclusion The expression of smad4 mRNA, TGFβ1 and TGFβR13 may be related to the occurrence, development, biological behavior and prognosis of gastric cancer, and may be used as an important biological marker.
