目的探讨维生素A在体内的活性产物视黄酸(RA)对1型糖尿病(T1DM)患儿外周血Th1/Th2平衡的影响。方法选取32例T1DM患儿为T1DM组,28例健康儿童为健康对照组。2组均在无菌条件下获取外周血单个核细胞(PBMC)悬液,均体外培养48 h。ELISA法检测培养上清液中IFN-γ、IL-4水平变化。采用SPSS11.5软件进行统计学分析。结果 1.T1DM组PBMC产生IFN-γ水平高于健康对照组(P<0.01),IFN-γ/IL-4比值明显高于健康对照组(P<0.01),IL-4水平与健康对照组比较差异无统计学意义(P>0.05);2.维生素A干预后T1DM组干预前PBMC产生IFN-γ水平明显低于(P<0.01),IFN-γ/IL-4比值明显低于T1DM组(P<0.01),IL-4水平高于T1DM组,但差异无统计学意义(P>0.05)。结论维生素A主要通过抑制Th1细胞功能,显著改善T1DM患儿Th1/Th2的失衡状态。
Objective To investigate the effect of retinoic acid (RA), the active product of vitamin A in the body, on the balance of Th1 / Th2 in children with type 1 diabetes mellitus (T1DM). Methods 32 cases of T1DM children were selected as T1DM group and 28 healthy children as healthy control group. Peripheral blood mononuclear cells (PBMCs) suspension were obtained under sterile conditions in both groups, and cultured in vitro for 48 h. The levels of IFN-γ and IL-4 in culture supernatants were detected by ELISA. Using SPSS11.5 software for statistical analysis. The level of IFN-γ in PBMC of T1DM group was higher than that in healthy control group (P <0.01), and the ratio of IFN-γ / IL-4 in PBMC of T1DM group was significantly higher than that in healthy control group (P> 0.05) .2. The level of IFN-γ in PBMC of T1DM group before intervention was lower than that of the control group (P <0.01), the ratio of IFN-γ / IL-4 was significantly lower than that of T1DM group (P <0.01). The level of IL-4 was higher than that of T1DM group, but the difference was not statistically significant (P> 0.05). Conclusion Vitamin A can significantly improve the imbalance of Th1 / Th2 in T1DM children by inhibiting the function of Th1 cells.
