目的:建立同时测定大鼠血浆中多索茶碱及其代谢产物茶碱的高效液相色谱法。方法:用18%高氯酸溶液处理血浆后,采用高效液相色谱法测定。色谱柱为Hypersil ODS C_(18)反相柱,流动相为甲醇-水(1:3),紫外检测波长271 nm,柱温为室温,流速l ml·min~(-1),进样量20μ1。结果:多索茶碱和茶碱的线性范围分别为0.5~50μg·ml~(-1)和0.2~20μg·ml~(-1)。日内和日间精密度RSD均小于9.6%。血浆样品稳定性符合要求。相对回收率97.02%~102.36%,绝对回收率大于85.0%。结论:本法专属性强,灵敏度高,重复性好,适用于多索茶碱的药物动力学研究。
Objective: To establish a HPLC method for the simultaneous determination of doxofylline and its metabolite theophylline in rat plasma. Methods: Plasma was treated with 18% perchloric acid solution and determined by high performance liquid chromatography. The chromatographic column was Hypersil ODS C_ (18) reversed-phase column. The mobile phase consisted of methanol-water (1: 3) and UV detection wavelength was 271 nm. The column temperature was at room temperature and the flow rate was 1 ml · min ~ 20μ1. Results: The linear ranges of doxofylline and theophylline were 0.5 ~ 50μg · ml ~ (-1) and 0.2 ~ 20μg · ml ~ (-1), respectively. Intra-day and inter-day precision RSDs were less than 9.6%. Plasma sample stability to meet the requirements. The relative recoveries were 97.02% ~ 102.36%, and the absolute recoveries were more than 85.0%. Conclusion: This method is specific, sensitive and reproducible, and is suitable for the pharmacokinetic study of doxofylline.
