目的:研究CD147和MMP-2在大肠癌组织中的表达和大肠癌患者血清中的水平变化,探讨其与大肠癌侵袭和转移的相关性,及二者间的相互联系。方法:应用免疫组织化学(SABC)法结合图像分析软件检测44例大肠癌、癌旁和正常对照组织中CD147和MMP-2的表达及半定量测定。应用酶联免疫吸附实验(ELISA)测定44例大肠癌患者血清中CD147和MMP-2的水平,并设36例健康对照。结果:大肠癌组织CD147和MMP-2的表达显著高于癌旁和正常对照组织,其表达与肿瘤分型、分化程度未见相关,而与转移密切相关。大肠癌患者血清CD147和MMP-2水平显著高于健康对照组,且伴转移组血清中CD147水平显著高于无转移组;低分化组中MMP-2水平显著高于高、中分化组。两者在组织定量和血清水平呈正相关。结论:大肠癌组织、血清中CD147高表达及高含量在一定程度上可以反映大肠癌发展程度及转移状态,其促MMP-2激活和表达增高的作用可能是其中一个重要因素。
OBJECTIVE: To investigate the expression of CD147 and MMP-2 in colorectal carcinoma and serum levels in colorectal cancer patients, and to explore its correlation with the invasion and metastasis of colorectal cancer and the relationship between them. Methods: Immunohistochemistry (SABC) combined with image analysis software was used to detect the expression of CD147 and MMP-2 in 44 cases of colorectal cancer, adjacent normal tissues and normal controls. Serum levels of CD147 and MMP-2 in 44 patients with colorectal cancer were measured by enzyme-linked immunosorbent assay (ELISA), and 36 healthy controls were also established. Results: The expressions of CD147 and MMP-2 in colorectal cancer tissues were significantly higher than those in para-cancerous tissues and normal control tissues. The expressions of CD147 and MMP-2 were not correlated with tumor type and differentiation, but closely correlated with metastasis. The levels of serum CD147 and MMP-2 in patients with colorectal cancer were significantly higher than those in healthy controls, and the serum level of CD147 in patients with metastasis was significantly higher than that in patients without metastasis. The levels of MMP-2 in patients with low-grade were significantly higher than those in patients with high-grade and moderate-grade. There was a positive correlation between tissue quantification and serum level. Conclusion: The high expression and high level of CD147 in colorectal cancer tissue and serum can reflect the development and metastasis of colorectal cancer to a certain extent. The effect of promoting the activation and expression of MMP-2 may be one of the important factors.
