本实验选用富含有血红蛋白特征面无核的正常小鼠网织红细胞,与小鼠淋巴瘤细胞株融合,对杂交细胞基因产物血红蛋白进行联苯胺组织化学染色、荧光抗体和生化测定。结果与电镜和流式细胞光度计的实验结果一致,表明珠蛋白基因产物血红蛋白住杂交细胞中,能够表达并成为融合细胞的标志。联苯胺阳性细胞数在HAT选择性培液筛选后数量增加。BW×R胞质杂交细肥在传代过程中,仍继续出现血红蛋白。文中对杂交细胞血红蛋白表达及其在传代过程中,在一部分细胞中丢失的可能机理进行了讨论。
In this experiment, normal mouse reticulocytes enriched with hemoglobin and non-nucleus were selected and fused with mouse lymphoma cell lines. The hemoglobin of hybrid cells was analyzed by benzidine histochemical staining, fluorescent antibody and biochemical assay. The results are consistent with the experimental results of electron microscopy and flow cytometry, indicating that the globin gene product hemoglobin in hybrid cells, can be expressed and become a fusion cell markers. The number of benzidine positive cells increased after screening by HAT selective culture. BW × R cytoplasm hybrid fine fertilizer in the passage of time, still continue to appear hemoglobin. The possible mechanism of hybrid cell hemoglobin expression and its loss in some cells during passage is discussed.
