目的建立一方便、实用和易于操作的轻度水肿性胰腺炎模型以更好地对急性胰腺炎的发病机制及防治措施进行研究。方法采用Wistar大鼠 ,以Caerulein作为急性水肿性胰腺炎的刺激诱导剂 ,将Caerulein预灌入一特制的渗透压泵 (Alzet)中 ,再将该泵植于大鼠左下腹 ,以静脉输入方式 ,将Caerulein由股静脉缓慢输入血液而诱发胰腺炎。结果由病理组织检查证实 ,以Caerulein(312.5mg/L,8μl/h)静脉灌注6h即显示明显组织水肿 ,大体检查胰腺各小叶间被清亮的胶冻样液体分隔 ,光镜检查显示胰腺间质水肿和明显的空泡化 ,但无炎症浸润 ;而仅以生理盐水灌注者则未见胰腺组织学的明显变化。结论采用渗透压泵植入大鼠腹部皮下以Caerulein缓慢静注能够可靠地诱导轻度水肿性胰腺炎 (组织学证实 ) ,该技术具有操作方便 ,效果可靠 ,并减少对动物干扰性操作的优点。
Objective To establish a convenient, practical and easy to operate model of mild edematous pancreatitis in order to better study the pathogenesis and prevention and treatment of acute pancreatitis. Methods Wistar rats were used Caerulein as stimulus inducer of acute edematous pancreatitis. Caerulein was pre-filled into a special osmotic pressure pump (Alzet), and then implanted into the left lower quadrant of the rats by intravenous infusion , The Caerulein slowly from the femoral vein into the blood induced pancreatitis. The results of pathological examination confirmed that Caerulein (312.5mg / L, 8μl / h) intravenous infusion of 6h showed significant tissue edema, the general examination of the pancreas was separated by clear jelly-like fluid between the lobular, light microscopy showed that the pancreatic stromal Edema and obvious vacuolization, but no infiltration of inflammation; while saline infusion alone was no significant change in pancreatic histology. CONCLUSIONS: Osteoporotic implants implanted subcutaneously in the abdomen of rats, slowly intravenously with Caerulein, can reliably induce mild edematous pancreatitis (histologically confirmed) with the advantages of easy handling, reliable results and reduced interference with animals .
