目的:选择杜仲叶醇提物中黄酮含量的测定方法。方法:分别采用紫外分光光度法、AlCl3显色法和Al(NO3)3络合法测定杜仲叶醇提液中黄酮的含量,并对测量结果进行了比较。结果:紫外分光光度法测得黄酮含量为4.4025mg/mL,数据偏高;络合法中,AlCl3显色法测定结果为0.2073mg/mL,低于Al(NO3)3络合法(0.4889mg/mL),甚至低于HPLC法测得的纯化后杜仲叶醇提液中黄酮含量(0.3201mg/mL)。结论:Al(NO3)3络合法测定结果较另外两种方法准确,是测定杜仲叶醇提液中总黄酮含量的有效方法。
Objective: To determine the content of flavonoids in ethanol extract of Eucommia ulmoides Oliv. Methods: The contents of flavonoids in alcohol extracts from leaves of Eucommia ulmoides Oliv. were determined by ultraviolet spectrophotometry, AlCl3 coloration method and Al(NO3)3 complex method. The results were compared. Results: The content of flavonoids was 4.4025mg/mL by ultraviolet spectrophotometry, and the data was high. In the complexation method, the result of AlCl3 coloration was 0.2073mg/mL, which was lower than that of Al(NO3)3 complex (0.4889mg (/mL), even lower than the HPLC method, the content of flavonoids in purified Eucommia ulmoides leaves (0.3201 mg/mL). Conclusion: The determination results of Al(NO3)3 complex method are more accurate than the other two methods. It is an effective method to determine the content of total flavonoids in alcohol extract of Eucommia ulmoides Oliv.
