目的:建立高效液相色谱-飞行时间质谱(LC-DAD-ESI-Q-TOF-MS)联用技术快速鉴定慈孝竹叶中的多种化学成分,为慈孝竹叶的开发与利用提供参考。方法:慈孝竹叶甲醇提取物经过Eclipse XD-C18(4.6 mm×150 mm,5μm)色谱柱,0.1%甲酸水-乙腈洗脱分离后,通过二极管阵列检测器采集紫外数据,最终结合负离子模式下获得的高分辨飞行时间质谱数据对化合物准分子离子峰与特征碎片离子分析,推测慈孝竹叶甲醇提取物的化学成分。结果:在45 min的色谱分离时间内,初步推测出慈孝竹叶中的30种化学成分,其中包括21种黄酮类成分,5种酚酸类成分,2种烯酸类成分,其中,在该植物中首次发现酚酸类、荭草素4’-O-己糖苷、去甲氧矢车菊黄素7-O-芸香糖苷等12个化学成分。结论:本研究建立了慈孝竹叶化学成分的快速鉴定方法,可为慈孝竹叶的开发与利用提供基础数据。
OBJECTIVE: To establish a rapid LC-DAD-ESI-Q-TOF-MS technique to identify a variety of chemical constituents in the leaves of CiXiao, and to provide a reference for the development and utilization of the leaves of CiXiao. Methods: The methanol extract of Chuxie philoxeroides leaves was separated on a column of Eclipse XD-C18 (4.6 mm × 150 mm, 5 μm) and eluted with 0.1% formic acid in water. The ultraviolet data were collected by a diode array detector and finally combined with negative ion mode Obtained by high-resolution time-of-flight mass spectrometry compound quasi-molecular ion peak and characteristic fragment ion analysis speculated that the chemical components of the methanol extract of Phyllostachys pubescens. Results: During the 45 min chromatographic separation time, 30 chemical constituents in the leaves of Chuxiong bamboo were preliminarily inferred, including 21 flavonoids, 5 phenolic acids and 2 alkenoic acids. Among them, For the first time, 12 chemical constituents such as phenolic acids, 4’-O-hexosidine and 4-O-rutinoside were found in plants. Conclusion: This study established a rapid identification method of chemical constituents in leaves of CiXiao, which provided the basic data for the development and utilization of CiXiao bamboo leaves.
