目的 研究晚期混合裂变产物内照射诱发Wistar大鼠外周血淋巴细胞次黄嘌呤鸟嘌呤磷酸核糖转移酶(HPRT)基因突变情况。方法 将晚期混合裂变产物自尾静脉注入 ,再用 5 -溴脱氧尿嘧啶 (BrdU)法测定HPRT基因的突变频率。结果 随着内照射剂量的增加 ,大鼠外周血淋巴细胞HPRT基因突变频率也不断上升 ,内照射剂量在 10 .178cSv以上的各组与对照组均有显著差异 (P <0 .0 1)。剂量效应关系符合线性模型。结论 BrdU法是一种快速、简便、较准确的测定放射性核素内照射损伤的方法 ,HPRT基因突变可作为一种很好的生物剂量计。
Objective To investigate the mutation of hypoxanthine guanine phosphoribosyltransferase (HPRT) gene in peripheral blood lymphocytes of Wistar rats induced by advanced mixed fission products. Methods Late mixed fission products were injected into the caudal vein, and then the mutation frequency of HPRT gene was determined by 5 - bromodeoxyuridine (BrdU) method. Results With the increase of internal dose, the frequency of HPRT gene mutation in peripheral blood lymphocytes was also increased. There was a significant difference (P <0.01) between the groups with the internal dose of 10178 cSv and the control group. The dose-response relationship is in line with the linear model. Conclusion BrdU method is a rapid, simple and accurate method for the determination of radionuclide irradiation injury. HPRT gene mutation can be used as a good biological dosimeter.
