目的:研究秦皮总香豆素对慢性高尿酸血症小鼠血尿酸水平的影响,并初步探讨其作用机理。方法:采用黄嘌呤600mg/kg(ig.)+氧嗪酸钾100mg/kg(sc.)建立小鼠慢性高尿酸血症模型,给予不同剂量秦皮总香豆素(440mg/kg、220mg/kg、110mg/kg),观察高尿酸血症小鼠血清尿酸、尿尿酸、黄嘌呤氧化酶、血清肌酐及尿素氮水平。结果:秦皮总香豆素组小鼠的血清尿酸及尿尿酸值明显低于模型组,有效抑制黄嘌呤氧化酶活性,血清肌酐及尿素氮值显著低于阳性对照组,均以大、中剂量组为佳。结论:秦皮总香豆素能明显降低高尿酸血症小鼠的血尿酸及尿尿酸水平,对肾功能损害较小,其机理可能与有效抑制黄嘌呤氧化酶活性有关。
OBJECTIVE: To study the effect of total coumarins of Qinpi on serum uric acid level in mice with chronic hyperuricemia and to investigate its mechanism. Methods: Chronic hyperuricemia model was induced in mice by xanthine 600mg / kg (ig.) + Oteracil 100mg / kg (sc.). The mice were given different doses of total coumarin (440mg / kg, 220mg / kg , 110mg / kg). The levels of serum uric acid, uric acid, xanthine oxidase, serum creatinine and blood urea nitrogen in hyperuricemic mice were observed. Results: The serum uric acid and uric acid of Qincundu total coumarin group were significantly lower than that of model group, and the inhibition of xanthine oxidase activity, serum creatinine and urea nitrogen were significantly lower than the positive control group, both in large and medium dose Good group. CONCLUSION: Total flavocoumarin of Qinpi can significantly reduce serum uric acid and uric acid level in mice with hyperuricemia and has less damage to renal function. The mechanism may be related to the inhibition of xanthine oxidase activity.
