目的筛选新疆紫草免疫活性有效部位。方法注射环磷酰胺(Cys)制备小鼠免疫低下模型,将小鼠随机分为空白对照组、模型组、阳性对照组及新疆紫草提取物高、中、低剂量治疗组,每组10只。空白对照组给予生理盐水,阳性对照组给予香菇菌多糖(4.5mg/kg),治疗组分别给新疆紫草石油醚、乙酸乙酯、正丁醇和水提取物,每天灌胃给药1次,连续9d。于给药第2天起,模型组、阳性对照组和治疗组同时腹腔注射Cys(50mg/kg),空白对照组注射生理盐水,连续3d。以碳粒廓清法测定非特异性免疫功能;血清溶血素法测定体液免疫功能;脾淋巴细胞增殖试验测定细胞免疫功能。结果新疆紫草水提物可使免疫功能低下小鼠的各项免疫指标均有不同程度的提高,分别为(模型/用药组)胸腺指数1.89/2.98,脾脏指数4.66/6.38,吞噬指数a 2.68/5.33、血清半数溶血值(HC50)62.5/88.4及脾淋巴细胞OD值0.35/0.6。结论新疆紫草水提物对免疫功能低下小鼠的非特异性免疫、体液免疫以及细胞免疫功能均有明显促进作用,为免疫活性有效部位。
Objective To screen the effective components of the immune activity of Lithospermum in Xinjiang. Methods Mice were immunosuppressed by injection of cyclophosphamide (Cys). The mice were randomly divided into blank control group, model group, positive control group and Xinjiang comfrey extract high, medium and low dose treatment group, 10 . The control group was given normal saline, and the positive control group was given polysaccharides of Lentinula edodes (4.5 mg / kg). The treatment groups were given petroleum ether, ethyl acetate, n-butanol and water extract of Xinjiang Lithospermum respectively, Continuous 9d. On the second day after administration, Cys (50mg / kg) was intraperitoneally injected into the model group, the positive control group and the treatment group, and the blank control group was injected with normal saline for 3d consecutive days. Determination of non-specific immune function by carbon clearance method; Determination of humoral immune function by hemolysin method; Measurement of cellular immune function by spleen lymphocyte proliferation test. The results of Xinjiang Arnebia water extract can make immunocompromised mice immune indicators have varying degrees of improvement, respectively (model / drug group) thymus index 1.89 / 2.98, spleen index 4.66 / 6.38, phagocytic index a 2.68 /5.33, half serum hemolysin (HC50) 62.5 / 88.4 and splenic lymphocyte OD 0.35 / 0.6. Conclusions Xinjiang Arnebia euchroma (Hance) extract can promote the nonspecific immunity, humoral immunity and cellular immune function in immunocompromised mice, which is an effective part of immune activity.
