目的探讨当归饮子对气血两虚型小鼠迟发型超敏变态反应的影响。方法 SPF级KM小鼠48只,其中40只采用利血平及乙酰苯肼制作动物气血两虚模型,随机分为模型组、氯雷他定组、当归饮子高剂量组、当归饮子中剂量组、当归饮子低剂量组,每组8只;另取8只相同标准小鼠作空白组。当归饮子高、中、低剂量组给予小鼠60、30、15 g/kg当归饮子加减方提取浓缩液灌胃,氯雷他定组给予1.66 mg/kg氯雷他定生理盐水溶液灌胃,模型组和空白组小鼠给予等体积生理盐水灌胃。每日1次,连续给药1周。除空白组外,每只小鼠腹部用1%DNCB溶液均匀涂抹进行致敏,致敏后第5天,将1%DNCB溶液均匀涂抹于小鼠右耳进行攻击,空白组同样涂耳但未致敏,攻击后24 h,测定每组小鼠耳廓肿胀度及抑制率。各组实验动物眼眶取血,采用双抗夹心ELISA法测试血清中Ig E水平。结果与空白组比较,模型组耳肿胀度明显增加,Ig E水平升高(P<0.01);与模型组比较,氯雷他定组及当归饮子3个剂量组耳肿胀度降低,Ig E水平降低(P<0.05,P<0.01),同时当归饮子高、中剂量组耳肿胀度降低优于氯雷他定组(P<0.05)。用药各组耳廓肿胀度抑制率从高至低依次为:当归饮子高剂量组67.3%,当归饮子中剂量组56.0%,当归饮子低剂量组48.1%,氯雷他定组47.3%。结论当归饮子方能抑制小鼠耳廓肿胀反应,降低血清中Ig E水平,对气血两虚证小鼠迟发型超敏反应有一定抑制作用。
Objective To investigate the effect of Angelica sinensis on delayed type hypersensitivity in qi and blood deficiency mice. Methods Forty-eight SPF KM mice were randomly divided into model group, loratadine group, high dose Angelica yinzi group, Angelica sinensis Middle dose group and Angelica sinensis low dose group, with 8 mice in each group; another 8 same standard mice were taken as blank group. Angelica high, medium and low dose groups were given mice 60,30,15 g / kg angelica Yin sub-extract concentrate extraction gavage, loratadine group given 1.66 mg / kg loratadine saline solution Gavage, model group and blank mice were given equal volume of normal saline. 1 day, continuous administration for 1 week. In addition to the blank group, each mouse abdomen with 1% DNCB solution smear evenly sensitized, 5 days after sensitization, the 1% DNCB solution evenly applied to the right mouse ear to attack the blank group also coated ears but not Sensitized, 24 h after challenge, the auricle swelling and inhibition rate of each group were measured. Blood was collected from orbital of experimental animals in each group, and serum IgE level was measured by double-antibody sandwich ELISA. Results Compared with the blank group, the degree of ear swelling in the model group increased significantly and the level of Ig E increased (P <0.01). Compared with the model group, the three groups of dose of loratadine and Angelica Yin decoction decreased the degree of ear swelling, IgE (P <0.05, P <0.01). In addition, the reduction of ear swelling in Angelica sinensis and medium dose group was better than loratadine group (P <0.05). The inhibition rates of auricle swelling degree in each group from high to low were as follows: Angelica sinensis high dose 67.3%, Angelica sinensis Yin dose 56.0%, Angelica sinensis low dose 48.1%, Loratadine 47.3% . Conclusion Angelica decoction can inhibit mouse ear auricle swelling reaction, reduce serum IgE levels, and have some inhibitory effects on delayed type hypersensitivity in qi and blood deficiency mice.
