目的:研究鸡冠花正丁醇萃取物对大鼠功能失调性子宫出血的改善作用,并探讨其作用机制。方法:将60只受孕SD大鼠随机分为空白组、模型组、宫血宁胶囊组(阳性对照,0.07 g/kg)和鸡冠花正丁醇萃取物高、中、低剂量组(4.32、2.16、1.08 g/kg),每组10只。除空白组外,其余各组大鼠均在妊娠第7天ig米非司酮溶液和米索前列醇溶液造成不完全流产,以复制功能失调性子宫出血模型。从妊娠第8天开始,给药组大鼠ig相应药物,空白组和模型组大鼠ig等体积生理盐水,每天早、晚各给药1次,连续7d。给药结束后,测定大鼠子宫出血量以及血清中血栓素(TXA_2)、前列环素(PGI_2)和肿瘤坏死因子α(TNF-α)含量。结果:与空白组比较,模型组大鼠子宫出血量显著增加(P<0.01),血清中TXA_2含量显著减少、PGI_2和TNF-α含量显著增加(P<0.01)。与模型组比较,各给药组大鼠的子宫出血量均显著减少、血清中TXA_2含量均显著增加(P<0.01);宫血宁胶囊组和鸡冠花正丁醇萃取物高剂量组大鼠血清中PGI_2、TNF-α含量,以及鸡冠花正丁醇萃取物中剂量组大鼠血清中TNF-α含量均显著减少(P<0.01)。结论:鸡冠花正丁醇萃取物对药物流产不全致大鼠功能失调性子宫出血有明显的改善作用,其作用机制可能与调节TXA_2/PGI_2动态平衡、抑制TNF-α的过度分泌有关。
Objective: To study the effect of n-butanol extract of celosia fargesii on dysfunctional uterine bleeding in rats and to explore its mechanism. Methods: Sixty pregnant SD rats were randomly divided into three groups: blank control group, model group, Gongxuening capsule group (positive control, 0.07 g / kg), and celiac nigrum extract high, medium and low dose groups (4.32, 2.16, 1.08 g / kg), 10 in each group. Except for the blank group, all other rats in the other groups were given incomplete abortion due to ig mifepristone solution and misoprostol solution on day 7 of pregnancy to reproduce the dysfunctional uterine bleeding model. From the 8th day of gestation, the rats in the drug-treated group were given ig corresponding drugs, blank group and model group rats with the same volume of normal saline. The rats were dosed once daily for 7 days. After administration, the amount of uterine bleeding and the contents of TXA 2, PGI 2 and TNF-α in serum were determined. Results: Compared with the blank group, the amount of uterine bleeding in the model group was significantly increased (P <0.01), the content of TXA_2 in the model group was significantly decreased, and the levels of PGI_2 and TNF-α were significantly increased (P <0.01). Compared with the model group, the uterine bleeding volume of rats in each administration group was significantly reduced, and the content of TXA_2 in serum was significantly increased (P <0.01). The rats in Gongxuening capsule group and n-butanol extract of high-dose group Serum levels of PGI_2 and TNF-α, as well as the content of TNF-α in the serum of normal butanol extract of celosia farina decreased significantly (P <0.01). CONCLUSION: The n-butanol extract of celosia farnesse can significantly improve dysfunctional uterine bleeding in rats induced by medical abortion. The mechanism may be related to the dynamic balance of TXA 2 / PGI 2 and the inhibition of TNF-α over-secretion.
