目的观察罗勒提取物对小鼠腹腔巨噬细胞(RAW264.7)生长及其胞内5-脂氧酶活性的影响。方法采用MTT比色法检测罗勒乙酸乙酯提取物或罗勒正丁醇提取物对正常RAW264.7细胞和经脂多糖刺激的RAW264.7细胞生长的影响,求出其IC50值;用罗勒乙酸乙酯提取物或罗勒正丁醇提取物分别干预经脂多糖处理的RAW264.7细胞,采用ELISA法测定并比较上清液中白三烯B4的质量浓度,求出其抑制率。结果罗勒乙酸乙酯提取物和罗勒正丁醇提取物对正常RAW264.7细胞的生长有不同程度的抑制作用,给药时间为24 h的IC50值分别为312.5μg/mL、350.9μg/mL,具有浓度依赖性;两种提取物对经脂多糖刺激的RAW264.7的生长有一定的增殖作用(P<0.05);罗勒乙酸乙酯提取物和罗勒正丁醇提取物对白三烯B4的最大抑制率分别为42.78%、38.63%,其中罗勒乙酸乙酯提取物的作用强于罗勒正丁醇提取物(P<0.05)。结论罗勒提取物体外对经脂多糖刺激的RAW264.7细胞生长的最大抑制质量浓度为100μg/mL,对细胞内5-脂氧酶活性具有抑制作用。
Objective To observe the effects of basil extract on the growth of mouse peritoneal macrophages (RAW264.7) and its intracellular 5-lipoxygenase activity. Methods MTT colorimetric assay was used to determine the effect of basil extract or basil butanol extract on the growth of RAW264.7 cells and lipopolysaccharide-stimulated RAW264.7 cells. IC50 value was calculated. Ester extract or n-butanol extract were respectively used to pretreat RAW264.7 cells treated with lipopolysaccharide. The inhibitory rate of leukotriene B4 was determined by ELISA and compared with the concentration of leukotriene B4 in supernatant. Results The ethyl acetate extract of basil and the extract of n-butanol from basil inhibited the growth of normal RAW264.7 cells to a certain extent. The IC50 values at 24 h were 312.5 μg / mL, 350.9 μg / mL, (P <0.05). The extract of basil and n-butanol extract of basil had the greatest effect on the growth of leukotriene B4 The inhibitory rates were 42.78% and 38.63%, respectively. The effect of basil ethyl acetate extract was stronger than that of basil butanol extract (P <0.05). Conclusion The maximum inhibitory concentration of basil extract on RAW264.7 cells stimulated by lipopolysaccharide is 100 μg / mL, which has an inhibitory effect on intracellular 5-lipoxygenase activity.
