目的探讨种子育麟散治疗不孕症的可能作用机制。方法选取30只SD雌性大鼠随机分为空白对照组、模型组、中药组,每组10只。模型组和中药组通过腹腔内注射丙氨瑞林、尿促性腺激素、注射用绒毛膜促性腺激素建立控制性超促排卵模型大鼠。造模开始的同时中药组给予种子育麟散7.2 g/(kg·d)灌胃,模型组、空白对照组给予等量生理盐水灌胃,均每日1次,每次2 ml,连续9天。于孕5天后分别采用免疫组化法、Western blot法检测大鼠子宫内膜整合素αvβ3、同源框(HOX)A10蛋白表达,RT-PCR法检测HOXA10、整合素αvβ3 mRNA表达。结果与空白对照组比较,模型组子宫内膜整合素αvβ3、HOXA10蛋白及HOXA10、整合素αvβ3 mRNA表达量明显减少(P<0.05);与模型组比较,中药组子宫内膜整合素αvβ3、HOXA10蛋白及整合素αvβ3、HOXA10 mRNA表达量明显增加(P<0.05)。结论种子育麟散能够提高大鼠子宫内膜整合素αvβ3、HOXA10蛋白及mRNA的表达,从而改善妊娠结局,可能是其治疗不孕症的机制之一。
Objective To explore the probable mechanism of seed Yulin scattered treatment of infertility. Methods Thirty SD female rats were randomly divided into blank control group, model group and TCM group, with 10 rats in each group. The model group and the traditional Chinese medicine group established controlled ovarian hyperstimulation model rats by intraperitoneal injection of ararelin, urinary gonadotropin and injection of chorionic gonadotropin. At the same time, the model group and the blank control group were given intragastric administration of 7.2 g / (kg · d) day. Immunohistochemistry and Western blot were used to detect the expressions of integrin αvβ3 and HOX A10 protein in rat endometrium respectively. The expression of HOXA10 and integrin αvβ3 mRNA was detected by RT-PCR. Results Compared with the blank control group, the expressions of integrin αvβ3, HOXA10, HOXA10 and integrin αvβ3 in the model group were significantly decreased (P <0.05). Compared with the model group, the levels of endometrial integrin αvβ3, HOXA10 Protein and integrin αvβ3, HOXA10 mRNA expression increased significantly (P <0.05). Conclusion Seed Yulin Decoction can improve the expression of endometrial integrin αvβ3, HOXA10 protein and mRNA in rats and improve the pregnancy outcome, which may be one of the mechanisms of infertility treatment.
