蛹虫草中含有多种生理活性物质,是中国名贵中药材之一。前期试验发现其发酵液中含有纤溶酶,具有开发成一种新型溶栓药物的价值。以试验室保存的8株蛹虫草为试验菌株,以纤溶酶酶活为指标,进行液体深层培养,确定以C-5作为诱变出发菌株,对其进行原生质体紫外诱变处理,筛选得到31株抗制霉素突变株,抗性菌株以纤溶酶酶活为指标进行液体深层发酵,得到8株纤溶酶产量高于出发菌株15.00%以上的突变株,并从中筛选出3株遗传稳定性良好的正突变株CY-8,CY-18,CY-25,其纤溶酶产量分别比出发菌株提高了28.58%,28.22%,21.78%。
Cordyceps militaris contains a variety of physiologically active substances, is one of China’s precious Chinese herbal medicines. Preliminary tests found that the fermentation broth contains plasmin, with the development of a new type of thrombolytic value. Eight strains of Cordyceps militaris preserved in the laboratory were used as experimental strains. Plasmin enzyme activity was taken as an index to conduct liquid deep culture. The strain C-5 was selected as the mutagenizing strain and subjected to protoplast UV mutagenesis. Thirty-one mutants resistant to nystatin and resistant strains were subjected to deep liquid fermentation using plasmin as an index. Eight mutants with plasmin higher than 15.00% were obtained and three strains were selected The stable positive mutant strains CY-8, CY-18 and CY-25 had 28.58%, 28.22% and 21.78% higher plasmin respectively than the original strain.
