目的:探讨外周血中Th1、Th2细胞及相关细胞因子在变应性鼻炎发病中的变化和作用机制。方法:以卵清蛋白致敏激发制成SD大鼠变应性鼻炎模型,以流式细胞仪检测外周血中Th1和Th2细胞的含量,采用酶联免疫吸附试验(ELISA)检测血清和鼻黏膜组织匀浆中的INF-γ,IL-4的含量。结果:与健康对照组相比,变应性鼻炎模型的Th1数量有下降趋势,但P>0.05,Th2细胞却明显升高,P<0.05。Th1/Th2显著偏离正常比值,P<0.01。血清和组织匀浆中INF-γ下降,P>0.05,IL-4含量明显升高,P<0.01。结论:大鼠变应性鼻炎模型外周血中Th2细胞及相关细胞因子的变化,在其发病过程中可能具有重要的意义。
Objective: To investigate the changes of peripheral blood Th1, Th2 cells and related cytokines in the pathogenesis of allergic rhinitis. Methods: The rat model of allergic rhinitis induced by ovalbumin challenge was established. The levels of Th1 and Th2 in peripheral blood were determined by flow cytometry. Serum and nasal mucosa were detected by enzyme-linked immunosorbent assay (ELISA) The content of INF-γ and IL-4 in tissue homogenate. Results: Compared with healthy control group, the number of Th1 in allergic rhinitis model had a decreasing trend, but P> 0.05, Th2 cells were significantly increased (P <0.05). Th1 / Th2 significantly deviated from the normal ratio, P <0.01. Serum and tissue homogenate INF-γ decreased, P> 0.05, IL-4 content was significantly increased, P <0.01. Conclusion: The changes of Th2 cells and related cytokines in the peripheral blood of rat allergic rhinitis model may play an important role in the pathogenesis of allergic rhinitis.
