目的探讨β-淀粉样蛋白(Aβ)对幼鼠骨髓干细胞增殖、迁移和凋亡的影响。方法培养新生小鼠骨髓干细胞,将其分为对照组和Aβ处理组。利用Brd U方法检测两组阳性细胞数;检测Aβ蛋白对幼鼠骨髓干细胞迁移的影响;流式细胞仪检测对照组和Aβ处理组细胞凋亡情况;Western印迹检测Cleaved-caspase-3、Bax、Bcl-2蛋白的表达。结果 Aβ处理组的细胞生长速度明显低于对照组,对照组Brd U阳性率显著高于Aβ处理组(P<0.05);Aβ处理组的细胞迁移距离及凋亡率显著高于对照组(P<0.05,P<0.01);经Aβ蛋白处理的骨髓干细胞中Cleaved-caspase-3、Bax蛋白表达量明显升高,而Bcl-2蛋白表达量与对照组相比明显减少(P<0.01)。结论 Aβ能抑制骨髓干细胞的增殖,加速骨髓干细胞的迁移和凋亡。
Objective To investigate the effects of β-amyloid protein (Aβ) on proliferation, migration and apoptosis of mouse bone marrow stem cells. Methods The newborn mouse bone marrow stem cells were cultured and divided into control group and Aβ treatment group. BrdU method was used to detect the number of positive cells in both groups; the effect of Aβprotein on the migration of bone marrow stem cells in young rats; the apoptosis of cells in control and Aβ treated groups were detected by flow cytometry; the expressions of Cleaved-caspase-3, Bax, Bcl-2 protein expression. Results The cell growth rate of Aβ-treated group was significantly lower than that of the control group. The positive rate of BrdU in control group was significantly higher than that of Aβ-treated group (P <0.05). The migration distance and apoptosis rate of Aβ-treated group were significantly higher than those of control group <0.05, P <0.01). The protein expression of Cleaved-caspase-3 and Bax in Aβ-treated bone marrow stem cells was significantly increased, while the expression of Bcl-2 protein was significantly decreased compared with the control group (P <0.01). Conclusion Aβ can inhibit the proliferation of bone marrow stem cells and accelerate the migration and apoptosis of bone marrow stem cells.
