目的:优选芪参健骨颗粒的纯化工艺。方法:采用单因素试验和L9(34)正交试验法,考察乙醇浓度、醇沉时间、药液密度等因素的影响,以黄芪甲苷、丹酚酸B的转移率为评价指标,多指标综合评分法(黄芪甲苷与丹酚酸B权重系数为0.6∶0.4)确定最佳纯化工艺。结果:芪参健骨颗粒的最佳纯化工艺为提取液减压浓缩至药液相对密度1.05~1.10,加95%乙醇使含醇量为50%,醇沉24 h。结论:优选的纯化工艺方法简便可行、工艺稳定、重复性好。
Objective: To optimize the purification process of Qishen Jiangu granules. Methods: The single factor test and L9 (34) orthogonal test were used to investigate the effects of ethanol concentration, alcohol precipitation time and drug solution density on the metastasis rate of astragaloside IV and salvianolic acid B, Comprehensive scoring method (Astragaloside and salvianolic acid B weight coefficient of 0.6: 0.4) to determine the best purification process. Results: The optimal purification process of Qishen Jiangu granules was as follows: the extract was concentrated under reduced pressure to a relative density of 1.05-1.10; 95% ethanol was added to make the alcohol content 50% and the alcohol precipitation 24 h. Conclusion: The preferred purification process is simple and feasible, the process is stable and reproducible.
