目的:探讨石上柏对人鼻咽癌TW03细胞的生长抑制作用及其可能的机制。方法:采用MTT法检测TW03细胞的生长抑制率,流式细胞仪检测TW03细胞周期改变,免疫组化法检测TW03细胞中Bcl-2和Bax的表达。结果:随着石上柏质量浓度的增加和作用时间的延长,TW03细胞生长抑制率显著升高(P<0.05)。随着石上柏质量浓度的增加,S期TW03细胞所占比例逐渐上升,而G_0/G_1期和G_2/M期所占比例逐渐下降,同时Bc1-2表达下调,而Bax表达上调。结论:石上柏能通过诱导人鼻咽癌TW03细胞周期阻滞于S期,其机制可能与下调Bcl-2表达和上调Bax的表达有关。
OBJECTIVE: To investigate the inhibitory effect of Shiseido on human nasopharyngeal carcinoma TW03 cells and its possible mechanism. Methods: The growth inhibition rate of TW03 cells was detected by MTT assay. The cell cycle of TW03 cells was detected by flow cytometry. The expression of Bcl-2 and Bax in TW03 cells was detected by immunohistochemistry. Results: The growth inhibition rate of TW03 cells was significantly increased with the increase of the mass concentration and the prolongation of action time (P <0.05). The proportion of TW03 cells in S phase increased gradually with the increase of the mass concentration of M. cypress, whereas the proportion of G0 / G1 phase and G_2 / M phase decreased gradually. Meanwhile, the expression of Bcl-2 was down-regulated and the expression of Bax was up-regulated. CONCLUSION: Shishangbai can induce the arrest of human nasopharyngeal carcinoma TW03 cell cycle in S phase by down-regulating the expression of Bcl-2 and up-regulating the expression of Bax.
