目的:研究膀胱平滑肌细胞与膀胱脱细胞基质(BAM)的生物相容性。方法:分离培养兔膀胱平滑肌细胞,采用α-平滑肌肌动蛋白(α-SMA)抗体进行鉴定。将以1×10n 5个/mL的单细胞悬液均匀接种于制备BAM支架上,通过与单独培养的膀胱平滑肌细胞作对照,绘制两组细胞生长曲线图,对比观察两条生长曲线的差异性。n 结果:第4代兔膀胱平滑肌细胞形态为典型的“长梭样”,免疫荧光显示α-SAM表达阳性。膀胱平滑肌细胞能够在BAM上很好地黏附生长,两组细胞生长曲线基本相似。结论:膀胱平滑肌细胞与BAM生物相容性好,可用膀胱平滑肌细胞作为种子细胞,以BAM作为支架材料构建组织工程化尿路移植物。“,”Objective:To study the biocompatibility of bladder smooth muscle cells and bladder acellular matrix(BAM).Methods:Bladder smooth muscle cells were isolated and cultured, their morphological characteristics were observed under inverted microscope, and identified by α -smooth muscle actin (α-SMA) antibody.Then the 4th generation bladder smooth muscle cells were seeded on BAM scaffold with a density of 1×10 n 5 cells /mL. The separated cultured bladder smooth muscle cells were regarded as control group. The cell growth curves of the two groups were drawn and the difference between them was observed.n Results:The morphology of the 4th generation bladder smooth muscle cells was typical “ long fusiform” . Immunofluorescence showed positive expression of α-SAM.Bladder smooth muscle cells could adhere and growth on the surface of BAM, and the cell growth curves of the two groups are basically similar.Conclusions:Bladder smooth muscle cells have good biocompatibility with BAM.Bladder smooth muscle cells can be used as seed cells and BAM can be used as scaffold material to construct tissue-engineered urinary tract graft.
