目的应用HPLC法测定大鼠血浆中木槿素A的质量浓度,对木槿素A在大鼠体内的药动学进行研究。方法采用Diamonsil C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数为0.1%甲酸水溶液(体积比79∶21),流速为1.0 m L·min-1,检测波长为228 nm,柱温为25℃。应用液-液萃取法处理血浆,测定6只大鼠静脉给予20 mg·kg-1木槿素A后不同时刻血浆中木槿素A的质量浓度,采用DAS2.1.1药动学软件计算药动学参数。结果木槿素A质量浓度在50~5 000μg·L-1内线性关系良好,日内、日间精密度均不大于9.2%。木槿素A的药动学参数如下:末端消除半衰期t1/2为0.884 h,末端消除速率ke为1.412 h-1。结论该方法操作简便,准确度高、灵敏度强、专属性好,可作为测定大鼠血浆中木槿素A质量浓度的方法,为探讨木槿素A的药动学行为提供方法依据,对于评价木槿素A疗效和安全性有重要意义。
OBJECTIVE To determine the concentration of hibitonin A in rat plasma by HPLC and to study the pharmacokinetics of hibisoid H in rats. Methods Diamonsil C18 column (250 mm × 4.6 mm, 5 μm) was used as the mobile phase. The mobile phase consisted of a methanol-water solution of 0.1% formic acid at a volume ratio of 79:21, a flow rate of 1.0 mL · min-1 and a detection wavelength of 228 nm, the column temperature is 25 ℃. The plasma was treated with liquid-liquid extraction method. The plasma concentrations of hyaluronidin A in 6 rats at different time points after intravenous injection of 20 mg · kg-1 Hibrinone-A were measured. The pharmacokinetic parameters were calculated by DAS2.1.1 pharmacokinetic software . Results The linearity of Hibiscus albican A was good in the concentration range of 50-5 000 μg · L-1, and the intra-day and inter-day precision was no more than 9.2%. The pharmacokinetic parameters of Hibiscusel A were as follows: the half-life of terminal elimination t1 / 2 was 0.884 h, and the terminal elimination rate ke was 1.412 h-1. Conclusions The method is simple, accurate, sensitive and specific. It can be used as a method to determine the concentration of hibrinosin A in rat plasma. It provides a method basis for exploring the pharmacokinetic behavior of hibiscus alnus A, A efficacy and safety of great significance.
