目的观察伽玛刀照射对致痫大鼠海马组织DBP及p-ERK1/2蛋白的影响,探讨伽玛刀治疗颞叶癫痫的作用机制。方法实验分正常对照组、模型组、伽玛刀治疗组,每组30只大鼠。以匹罗卡品腹腔注射复制大鼠颞叶癫痫模型,实验设立伽玛刀治疗后3h、7d、15d、30d、60d等五个不同时间点进行指标检测,应用western blotting法检测大鼠海马组织DBP、p-ERK1/2蛋白的表达。结果匹罗卡品点燃模型后大鼠海马的DBP被显著激活,并随时间出现有规律的变化,在各时间点模型组大鼠海马区DBP蛋白的表达与正常对照组相比具有统计学差异(P<0.05),而伽玛刀放射治疗能显著抑制这种激活(P<0.05)。海马区p-ERK1/2蛋白的变化呈现与DBP蛋白相同的规律。结论伽玛刀对DBP-ERK1/2信号通路的影响可能是其治疗颞叶癫痫的作用机制之一。
Objective To observe the effect of gamma knife irradiation on the expression of DBP and p-ERK1 / 2 protein in hippocampus of epileptic rats and to explore the mechanism of gamma knife treatment of temporal lobe epilepsy. Methods The experiment was divided into normal control group, model group and gamma knife treatment group, 30 rats in each group. The rat models of temporal lobe epilepsy were injected intraperitoneally with pilocarpine. The rats were treated with Gamma Knife for 3h, 7d, 15d, 30d, 60d and other five time points to detect the indexes. The hippocampal tissues were detected by western blotting DBP, p-ERK1 / 2 protein expression. Results DBP was significantly activated in the hippocampus of rats exposed to pilocarpine and showed regular changes with time. At each time point, the expression of DBP in the hippocampus of rats in the model group was significantly lower than that of the normal control (P <0.05), while gamma knife radiotherapy significantly inhibited this activation (P <0.05). The change of p-ERK1 / 2 protein in hippocampus showed the same pattern as DBP protein. Conclusion The effect of Gamma Knife on DBP-ERK1 / 2 signal pathway may be one of its mechanisms of treatment for temporal lobe epilepsy.
