目的探讨人参多糖对冷应激大鼠生殖功能的影响,研究大鼠睾丸组织雄激素受体mRNA表达与血浆睾酮变化及人参多糖的调节作用。方法采用大鼠雄激素受体(AR)cDNA探针斑点杂交法检测雄激素受体mRNA表达强度,血浆睾酮含量测定采用放射免疫分析法。结果对照组睾酮含量为(10.93±1.44)nmol.L-1,冷应激实验组为(6.06±1.78)nmol.L-1,冷应激人参多糖实验组为(8.01±1.38)nmol.L-1。实验组与对照组比较,差异显著(P<0.05)。Dot?blot杂交结果:冷应激实验组明显弱于对照组,冷应激加人参多糖实验组弱于对照组但强于冷应激实验组。结论冷应激使睾酮水平下降,雄激素受体mRNA表达信号减弱,人参多糖可促进冷应激条件下的睾酮生成,加强雄激素受体mRNA表达。
Objective To investigate the effects of ginseng polysaccharides on the reproductive function of cold-stressed rats, to study the changes of androgen receptor mRNA expression and plasma testosterone levels in the testis of rats and the regulatory effects of ginseng polysaccharides. Methods The expression of androgen receptor mRNA was detected by dot-blot hybridization with the rat androgen receptor (AR) cDNA probe. The plasma testosterone content was determined by radioimmunoassay. Results The content of testosterone in the control group was (10.93±1.44)nmol.L-1, in the cold stress experiment group was (6.06±1.78) nmol.L-1, and in the cold stress ginseng polysaccharide group was (8.01±1.38) nmol.L. -1. The difference between the experimental group and the control group was significant (P<0.05). Dot-blot hybridization results: The cold stress experimental group was significantly weaker than the control group, and the cold stress plus ginseng polysaccharide experimental group was weaker than the control group but stronger than the cold stress experimental group. Conclusions Cold stress reduced the level of testosterone and decreased the signal of androgen receptor mRNA expression. Ginseng polysaccharides can promote the production of testosterone under cold stress condition and strengthen the expression of androgen receptor mRNA.
