目的探讨胚胎培养液中可溶性人白细胞抗原G(sHLA-G)与胚胎发育以及IVF-ET结局的关系,为临床优选胚胎提供一种有效的非侵入性检测途径。方法采用酶联免疫吸附试验(ELISA)检测第3日移植胚胎培养液中sHLA-G的表达,根据移植胚胎培养液中sHLA-G的表达分组比较体外受精-胚胎移植的结局。结果共收集胚胎培养液284份,sHLA-G阳性率为55.28%(157/284)。根据胚胎培养液sHLA-G的检出分组比较,sHLA-G++组和sHLA-G+-组的优质胚胎率均明显高于sHLA-G--组(P<0.05),sHLA-G++组和sHLA-G+-组的临床妊娠率均高于sHLA-G--组(P<0.05),sHLA-G++组与sHLA-G+-组间的优质胚胎率和临床妊娠率均无统计学差异(P>0.05)。结论 sHLA-G在早期胚胎培养液中有表达,能反映了胚胎良好的发育和种植潜能,预示着较好的IVF-ET结局,是一种优选移植胚胎的有效的非侵入性途径。
Objective To investigate the relationship between soluble human leukocyte antigen G (sHLA-G) and embryonic development and IVF-ET outcome in embryo culture medium and to provide an effective and noninvasive detection method for clinically preferred embryos. Methods The expression of sHLA-G was detected by enzyme-linked immunosorbent assay (ELISA) in embryo culture fluid of day 3, and the results of in vitro fertilization-embryo transfer were compared according to the expression of sHLA-G in the transplanted embryo culture fluid. Results A total of 284 embryos were collected. The positive rate of sHLA-G was 55.28% (157/284). According to the detection group of sHLA-G in embryo culture medium, the high-quality embryos of sHLA-G ++ group and sHLA-G + - group were significantly higher than that of sHLA-G-- group (P <0.05) The clinical pregnancy rates of G + -group were higher than that of sHLA-G-group (P <0.05), but there was no significant difference between high-quality embryos and clinical pregnancy rates between sHLA-G ++ group and sHLA-G + ). Conclusion The expression of sHLA-G in early embryo culture medium can reflect the good development and implantation potential of embryos, which indicates the better outcome of IVF-ET. It is an effective and noninvasive method for transplanted embryos.
