目的:探求一种合理有效的研究中药作用于中枢神经系统药效的方法。方法:利用体外细胞培养的方法,以三种不同浓度观察血清、中药血清、脑脊液、中药脑脊液对于星形胶质细胞和神经元的影响。结果:台盼兰染色和LDH检测证明正常血清和中药血清在三种浓度均出现对细胞的毒害作用(P<0.05),尤以100μl、200μl为重,且在同一浓度时,两组之间没有明显的差异(P>0.05)。而正常脑脊液组和中药脑脊液组与空白对照组相比,未见明显差异(P>0.05);MTT检测证明中药脑脊液组OD值明显高于其余各组(P<0.05)。结论:利用脑脊液进行中药对中枢神经系统的药效观察明显优于血清,提出“脑脊液药理学”的概念;中药可诱导星形胶质细胞产生神经营养作用。
Objective: To explore a reasonable and effective method to study the effect of Chinese medicine on the efficacy of central nervous system. METHODS: Using in vitro cell culture methods, the effects of serum, Chinese herbal serum, cerebrospinal fluid, and cerebrospinal fluid on astrocytes and neurons were observed at three different concentrations. RESULTS: Trypan blue staining and LDH assay demonstrated that normal serum and traditional Chinese medicine serum had toxic effects on cells in all three concentrations (P<0.05), especially in 100 μl and 200 μl, and at the same concentration, between the two groups. There was no significant difference (P>0.05). The normal cerebrospinal fluid group and the Chinese medicine cerebrospinal fluid group had no significant difference compared with the blank control group (P>0.05). The MTT test proved that the OD value of the Chinese medicine cerebrospinal fluid group was significantly higher than the other groups (P<0.05). Conclusion: The therapeutic effect of CSF on Chinese central nervous system is superior to that of serum. The concept of cerebrospinal fluid pharmacology is proposed. Chinese medicine can induce neurotrophic effect of astrocytes.
