目的探讨基质金属蛋白酶-9(MMP-9)与孕妇子痫前期发病的相关性。方法将2013年4月-2014年6月在该院住院分娩的孕妇按照其妊娠20周以后的血压以及蛋白尿情况分为实验组(子痫前期66例)和对照组(正常妊娠34例),实验组分为轻度子痫前期组(33例)和重度子痫前期组(33例),应用免疫组化SP法对两组胎盘组织中MMP-9的表达水平进行检测,并测定其光密度值。同时对每个新生儿体重和胎盘重量进行测量和记录。结果在胎盘组织中,MMP-9的光密度值实验组低于对照组(t=4.375,P<0.001),轻度子痫前期组高于重度子痫前期组(t=5.302,P<0.001)。实验组胎盘重量(t=3.181,P=0.002)和新生儿体重(t=3.034,P=0.003)低于对照组,轻度子痫前期组胎盘重量(t=5.128,P<0.001)和新生儿体重(t=4.956,P<0.001)均高于重度子痫前期组。在胎盘组织中,MMP-9的表达水平与胎盘重量及新生儿体重呈正相关。与新生儿体重<2.5 kg者相比,≥2.5 kg者的MMP-9在胚胎组织中的表达水平较高(t=17.121,P<0.001)。结论 MMP-9在孕妇子痫前期的胎盘组织中表达程度低,孕妇病情恶化使其表达程度逐渐降低,表明MMP-9可能和子痫前期的发病有关。
Objective To investigate the relationship between matrix metalloproteinase-9 (MMP-9) and preeclampsia in pregnant women. Methods Pregnant women hospitalized in this hospital from April 2013 to June 2014 were divided into experimental group (preeclampsia 66 cases) and control group (normal pregnancy 34 cases) according to their blood pressure and proteinuria after 20 weeks of gestation. , The experimental group was divided into mild preeclampsia group (33 cases) and severe preeclampsia group (33 cases). The expression of MMP-9 in placenta was detected by immunohistochemical SP method, Optical density value. At the same time, each newborn weight and placental weight were measured and recorded. Results In the placenta, the optical density of MMP-9 was significantly lower in experimental group than in control group (t = 4.375, P <0.001), mild preeclampsia group was higher than that in severe preeclampsia group (t = 5.302, P <0.001) ). The placental weight (t = 3.181, P = 0.002) and neonatal body weight (t = 3.034, P = 0.003) in the experimental group were significantly lower than those in the control group Body weight (t = 4.956, P <0.001) were higher than those in severe preeclampsia group. In placental tissue, the expression level of MMP-9 positively correlated with the weight of the placenta and the weight of the newborn. Compared with neonates weighing <2.5 kg, MMP-9 levels of ≥ 2.5 kg were higher in embryonic tissues (t = 17.121, P <0.001). Conclusion The expression of MMP-9 in placental tissue of pregnant women with preeclampsia is low, and the expression of MMP-9 is gradually decreased, which indicates that MMP-9 may be related to the pathogenesis of preeclampsia.
