目的 研究 VP- 1 6诱导 PC- 3细胞凋亡和癌基因 bcl- 2及 C- myc表达的关系。方法 用末端标记法(TUNEL )和流式细胞术 (FCM)研究 PC- 3细胞凋亡 ,以免疫组化方法研究 bcl- 2及 C- myc蛋白的表达。结果 TUNEL和FCM检测表明 ,PC- 3细胞的凋亡率随 VP- 1 6作用浓度增加和作用时间延长而升高 ;免疫组化结果显示 :bcl- 2和 c- myc基因表达的阳性百分率随 VP- 1 6作用浓度增加和作用时间延长而降低。结论 VP- 1 6能够诱导 PC- 3细胞凋亡和抑制其bcl- 2和 c- myc基因的表达 ,且具有作用浓度和作用时间依赖性。
Objective To investigate the relationship between the apoptosis of PC-3 cells induced by VP-1 6 and the expression of oncogenes bcl-2 and C-myc. Methods The apoptosis of PC-3 cells was studied by TUNEL and flow cytometry (FCM). The expressions of bcl-2 and C-myc were studied by immunohistochemistry. Results The results of TUNEL and FCM showed that the apoptosis rate of PC-3 cells increased with the increase of concentration of VP-1 6 and the effect of prolonged time. Immunohistochemistry showed that the positive percentage of bcl-2 and c-myc VP-1 6 concentration increased and the role of prolonged decrease. Conclusion VP-1 6 can induce the apoptosis of PC-3 cells and inhibit the expression of bcl-2 and c-myc genes in a dose-and time-dependent manner.
