目的:本研究应用链脲佐菌素(STZ)腹腔注射制备糖尿病胃轻瘫(diabetic gastroparesis,DGP)大鼠模型,给予中药麻枳化浊方及西沙必利对该模型进行干预治疗,放免法测定大鼠胃动素(Motilin,MTL)、胃泌素(Gastrin,GAS)等胃肠激素的含量,通过观察大鼠的一般情况、MTL、GAS激素含量表达水平,从而研究探讨麻枳化浊方对糖尿病胃轻瘫大鼠MTL、GAS含量的影响,探讨麻枳化浊方对DGP大鼠胃肠的作用。方法:将40只健康雄性wistar大鼠随机分为5组:正常组、模型组、中药组、西药组、联合组,药物治疗组分别给予麻枳化浊方、西沙必利、及西沙必利加麻枳化浊方,模型组和正常组给予同等体积的生理盐水灌胃,应用放免法测定大鼠MTL、GAS等激素含量的表达水平。结果:实验大鼠的一般状况,正常组大鼠精神状态良好,活动正常,饮水、进食量、尿量无明显改变,体重逐渐增加。模型组于造模后出现多饮、多食、多尿,此后一直持续,体重未见增长,皮毛干枯毛躁泛黄。中药组、西药组、联合组等大鼠的一般情况较模型组有一定的改善。在试验过程中,模型组大鼠由于造模不当死亡一只。各组大鼠MTL含量的表达水平:中药组、西药组及联合组MTL含量明显低于模型组,差异有统计学意义(P<0.05),三者之间无明显差别(P>0.05),联合组与正常组之间无显著性差异(P>0.05)。各组大鼠GAS含量的表达水平:与模型组相比,中药组、西药组、联合组GAS含量明显降低(P<0.05),其中中药组与西药组之间无明显差异(P>0.05);联合组较中药组、西药组GAS含量减低,差异有统计学意义(P<0.05),与正常组相比差异无明显统计学意义(P>0.05)。结论:麻枳化浊方可能通过改善DGP大鼠MTL、GAS等胃肠激素的水平治疗DGP,为中医药治疗DGP提供了实验依据。
OBJECTIVE: In this study, diabetic rat models of diabetic gastroparesis (DGP) were established by intraperitoneal injection of streptozotocin (STZ), treated with traditional Chinese medicine Mazhuru Huazhuo Formula and cisapride, The levels of motilin (MTL), gastrin (GAS) and other gut hormones in rats were determined. The general situation of rats, MTL, GAS expression levels were studied, Fang on diabetic gastroparesis in rats MTL, GAS content, to explore Mazhuru Huazhuo Fang DGP rats gastrointestinal effects. Methods: Forty healthy male wistar rats were randomly divided into five groups: normal group, model group, traditional Chinese medicine group, western medicine group, combined group, and drug treatment group were given Mazhuthuazhuo Fang, cisapride, and cisapride Ma Zhuru Huazhuo Fang, model group and normal group were given the same volume of physiological saline gavage, the application of radioimmunoassay MTL, GAS and other hormone levels in rats. Results: The general condition of experimental rats, normal group, mental state is good, normal activities, drinking water, food intake, urine volume did not change significantly, weight increased gradually. The model group showed more drink, more food and more urine after the model was established. Chinese medicine group, Western medicine group, combined group and other rats in general than the model group has some improvement. During the experiment, the model group died of improper modeling. The MTL content of rats in each group was significantly lower than that in the model group (P <0.05), but there was no significant difference among the three groups (P> 0.05) There was no significant difference between the combined group and the normal group (P> 0.05). Compared with the model group, the GAS content of the Chinese medicine group, the Western medicine group and the combined group was significantly lower (P <0.05), and there was no significant difference between the Chinese medicine group and the western medicine group (P> 0.05) ; Compared with the traditional Chinese medicine group and Western medicine group, the GAS content decreased in the combined group (P <0.05). There was no significant difference between the combined group and the normal group (P> 0.05). Conclusion: Mazhuhuazhuo prescription may improve the DGP rat MTL, GAS and other gastrointestinal hormones in the treatment of DGP, providing experimental evidence for the treatment of DGP.
