目的:总结间质成纤维细胞与乳腺癌发生发展的研究现状,探讨间质成纤维细胞在乳腺癌发生、生长和转移中的作用及对治疗的影响。方法:应用Medline、PubMed及CNKI期刊全文数据库检索系统,以“乳腺癌、间质、肿瘤相关成纤维细胞、肿瘤微环境”为中文关键词,“breast cancer、stromal、carcinoma-associated fibroblasts、tumor microenviron-ment”等为英文关键词,检索1994-2011年的相关文献,共检索到英文文献258条,中文文献56条。纳入标准:1)乳腺癌间质成纤维细胞的形态和特性;2)分离和培养的方法;3)功能的研究;4)对乳腺癌的发生、生长、转移,以及治疗的影响。结果:乳腺癌间质成纤维细胞通过细胞与细胞间的相互接触,在各种趋化因子和(或)细胞因子的作用下,通过不同的信号通路促进上皮细胞恶性转化,在乳腺癌的发生、生长及转移中起着重要的作用,但具体的机制尚不明确。结论:乳腺癌间质成纤维细胞通过细胞间的相互作用,促进细胞的恶性转化,并通过分泌各种的细胞因子使得乳腺癌细胞的增殖、转移和侵袭能力大大加强,促进了乳腺癌的发展。它对于乳腺癌的内分泌治疗和化疗疗效有着重要的影响,可能是乳腺癌治疗的新靶点。
OBJECTIVE: To summarize the current situation of the development of mesenchymal fibroblasts and breast cancer, and to explore the role of mesenchymal fibroblasts in the occurrence, growth and metastasis of breast cancer and the impact on the treatment. Methods: Using the full-text database retrieval system of Medline, PubMed and CNKI, the key words of “breast cancer, stroma, tumor-associated fibroblasts, tumor microenvironment” and “breast cancer, stromal, carcinoma- associated fibroblasts , Tumor microenviron-ment ”and other English keywords, retrieved the relevant documents from 1994 to 2011, retrieved a total of 258 English articles and 56 Chinese articles. Inclusion criteria: 1) morphology and characteristics of breast mesenchymal fibroblasts; 2) methods of isolation and culture; 3) functional studies; and 4) effects on the occurrence, growth, metastasis, and treatment of breast cancer. Results: Breast cancer mesenchymal fibroblasts can promote the malignant transformation of epithelial cells through different signaling pathways by the interaction of cells and cells under the action of various chemokines and / or cytokines. In the occurrence of breast cancer , Growth and metastasis play an important role, but the specific mechanism is not yet clear. Conclusion: The breast mesenchymal fibroblasts can promote the malignant transformation of cells through the cell-cell interaction and enhance the proliferation, metastasis and invasion ability of breast cancer cells through the secretion of various cytokines and promote the development of breast cancer . It has an important effect on the endocrine and chemotherapy effects of breast cancer and may be a new target for the treatment of breast cancer.
