目的:观察硫酸锌对脑缺血再灌注损伤的作用。方法:用Pulsineli四动脉结扎法造成大鼠急性前脑缺血40min后再灌注1h的损伤模型,同时应用Fura-2/AM荧光指示剂测定硫酸锌对胎鼠脑细胞内游离钙浓度的影响。结果:静脉注射硫酸锌10mg·kg-1可降低钙在脑细胞内累积及丙二醛含量升高;减轻脑水肿并缓解脑损伤引起的LDH释放;改善EEG的抑制状态。在胎鼠脑细胞悬液中,硫酸锌(100~300μmol·L-1)可抑制高钾和L-谷氨酸引起的细胞内钙升高。结论:硫酸锌对急性再灌注脑损伤有保护作用,此作用与其防止钙超载、抗脂质过氧化物产生有关。
Objective: To observe the effect of zinc sulfate on cerebral ischemia-reperfusion injury. Methods: The model of injury was induced by Pulsineli four-artery ligation for 40 min after reperfusion in rats with acute forebrain ischemia. Fura-2 / AM fluorescent indicator was used to determine the effect of zinc sulfate on intracellular free calcium concentration in fetal rat brain. Results: Intravenous injection of zinc sulfate 10 mg · kg-1 could reduce the accumulation of calcium in brain cells and increase the content of malondialdehyde; relieve cerebral edema and relieve the release of LDH induced by brain injury; and improve the inhibition of EEG. In fetal rat brain cell suspension, zinc sulfate (100-300μmol·L-1) inhibited intracellular calcium elevation caused by high potassium and L-glutamate. Conclusion: Zinc sulphate has a protective effect on brain injury induced by acute reperfusion, and its effect is related to its prevention of calcium overload and anti-lipid peroxidation.
