目的探讨分泌性卷曲相关蛋白1(sFRP-1)和Wnt-1蛋白在宫颈上皮内瘤变(CIN)和宫颈癌中的表达及意义。方法采用免疫组化SABC法测定sFRP-1、Wnt-1蛋白在正常宫颈组织、低度上皮内瘤变组织(LSIL)、高度上皮内瘤变组织(HSIL)和宫颈癌中的表达水平及分布;并用反转录聚合酶链反应(RT-PCR)检测sFRP-1、Wnt-1 mRNA的表达水平。结果sFRP-1在宫颈癌中的表达明显低于HSIL及LSIL,同时LSIL低于正常宫颈组织。Wnt-1在宫颈癌中的表达明显高于HSIL及LSIL,同时HSIL高于LSIL,LSIL高于正常宫颈组织。结论Wnt信号通路抑制因子sFRP-1的过低表达及其主要成员Wnt-1的过度表达在由CIN发展到宫颈癌的过程中起了促进作用。
Objective To investigate the expression and significance of sFRP-1 and Wnt-1 in cervical intraepithelial neoplasia (CIN) and cervical cancer. Methods The expression and distribution of sFRP-1 and Wnt-1 protein in normal cervical tissue, low grade intraepithelial neoplasia (LSIL), high grade intraepithelial neoplasia (HSIL) and cervical cancer were determined by immunohistochemical SABC method. ; And sFRP-1, Wnt-1 mRNA expression levels were detected by reverse transcription-polymerase chain reaction (RT-PCR). Results The expression of sFRP-1 in cervical cancer was significantly lower than that in HSIL and LSIL, while LSIL was lower than that in normal cervical tissue. Wnt-1 expression in cervical cancer was significantly higher than HSIL and LSIL, while HSIL higher than LSIL, LSIL higher than normal cervical tissue. Conclusion The over-expression of Wnt signaling inhibitor sFRP-1 and the over-expression of Wnt-1, a major member of Wnt signaling pathway, play an important role in the progression from CIN to cervical cancer.
